摘要
研究民族药羊屎条Viburnum utile乙醇提取物的化学成分。采用D101大孔树脂、硅胶、Sephadex LH-20柱色谱等手段进行分离,利用NMR、MS等波谱技术鉴定结构并开展α-葡萄糖苷酶抑制活性检测。从该药材90%乙醇提取物中共分离16个化合物,分别鉴定为viburnumide A(1)、维生素E(2)、α-香树脂醇(3)、α-托可醌(4)、narjatamanin B(5)、咖啡酸乙酯(6)、柚皮素(7)、芹菜素(8)、ethylβ-D-ribo-hex-3-ulopyranoside(9)、反式对羟基肉桂酸(10)、穗花杉双黄酮(11)、芹菜素-7-O-β-D-葡萄糖苷(12)、木犀草素-7-O-β-D-葡萄糖苷(13)、鄂西香茶菜苷(14)、熊果酸(15)、2α-羟基齐墩果酸(16)。其中化合物1为新化合物,化合物2~16均为首次从羊屎条中分离得到。化合物11的α-葡萄糖苷酶抑制作用(IC50为4.0μmol·L^(-1))与阳性对照阿卡波糖(IC50为2.3μmol·L^(-1))相当。
The chemical constituents in the ethanol extract of Viburnum utile were isolated by column chromatography with D101 macroporous resin,silica gel,and Sephadex LH-20.Their structures were identified by spectroscopic techniques such as NMR and MS.All the compounds were evaluated for the inhibitory activities againstα-glucosidase.Sixteen compounds were obtained and identified as viburnumide A(1),vitamin E(2),α-amyrin(3),α-tocoquinone(4),narjatamanin B(5),ethyl caffeate(6),naringenin(7),apigenin(8),ethylβ-D-ribo-hex-3-ulopyranoside(9),trans-p-hydroxycinnamic acid(10),amentoflavone(11),apigenin-7-O-β-D-glucoside(12),luteolin-7-O-β-D-glucoside(13),henryoside(14),ursolic acid(15),and 2α-hydroxy-oleanolic acid(16).Among them,compound 1 was a new compound,and compounds 2-16 were isolated from V.utile for the first time.The inhibitory activity(IC50 of 4.0μmol·L^(-1))of compound 11 againstα-glucosidase was comparable to that of the positive control acarbose(IC50 of 2.3μmol·L^(-1)).
