摘要
N6-甲基腺苷(N6-methyladensine,m6A)修饰在细胞分化、免疫和肿瘤发生发展中具有重要作用,也是近几年表观转录组学的研究热点。甲基转移酶3(methyltransferase-like 3,METTL3)是m6A甲基转移酶复合体(methyltransferase complex,MTC)中的唯一催化亚基,其介导的m6A修饰在胃癌(gastric cancer,GC)发生发展中起着重要作用,因此可能成为新的肿瘤表观遗传调节剂。该文系统总结METTL3的结构基础、生物学功能,并阐述了METTL3在胃癌中的致癌功能及其潜在分子机制,以期为胃癌的治疗提供新的思路及方案。
N6-methyladensine(m6A)modification plays an important role in cell differentiation,immunity and tumorigenesis and development,and has been the focus of epigenetic transcriptomics research in recent years.Methyltransferase-like 3(METTL3)is the only catalytic subunit in the m6A methyltransferase complex(MTC),and its mediated m6A modification plays an important role in the occurrence and development of gastric cancer(GC).Therefore,it may become a new epigenetic modulator of tumor.In this paper,the structural basis and biological function of METTL3 are systematically summarized,and the carcinogenic function and potential molecular mechanism of METTL3 in gastric cancer are expounded,in order to provide new ideas and schemes for the treatment of gastric cancer.
