玉米多糖对阿霉素诱导心肌炎症损伤的改善作用及机制研究

摘要 探讨玉米多糖(corn polysaccharide,CPA)对阿霉素(doxorubicin,DOX)诱导心肌炎症损伤的作用及其机制。腹腔注射DOX制备C57小鼠急性心肌损伤模型,心脏超声检测小鼠心功能,记录心脏与机体质量之比,测定血清中心肌酶和抗氧化酶活性,HE染色观察心脏病理变化。采用MTT法测定CPA对DOX致H9c2心肌细胞的存活率,采用ELISA测定心肌组织和细胞中TNF-α、IL-6和IL-10的水平,采用Western blot检测NLRP3和Cleaved caspase-1蛋白含量。结果表明:与正常组相比,DOX组心肌酶升高,心肌组织损伤明显;与DOX组相比,CPA+DOX组心肌组织损伤程度较低。DOX组H9c2心肌细胞存活率降低,CPA+DOX组抗炎细胞因子较DOX组明显升高,组织及细胞中NLRP3、caspase-1和IL-1β水平切割蛋白减少。由此得知CPA在体内外实验中均可改善DOX所致心脏毒性的影响。 In order to investigate the effects and mechanisms of corn polysaccharide(CPA)on doxorubicin(DOX)-induced myocardial inflammatory damage,acute myocardial injury models in C57 mice were prepared through intraperitoneal injection of DOX.Mice cardiac function was detected by echocardiography,and the ratio of heart weight to body weight was recorded.Serum levels of cardiac enzymes and antioxidant enzyme activities were measured,and myocardial tissue changes were observed using HE staining.The MTT method was utilized to determine the viability of H9c2 cardiomyocytes treated with CPA.ELISA was used to measure the levels of TNF-α,IL-6,and IL-10 in myocardial tissue and cells.Western blot was used to detect the protein levels of NLRP3 and Cleaved caspase-1.The results showed that compared to the normal group,the DOX group exhibited elevated cardiac enzymes and noticeable myocardial tissue damage.The CPA+DOX group showed lower levels of myocardial tissue damage compared to the DOX group.The DOX group displayed a reduced survival rate of H9c2 myocardial cells,while the CPA+DOX group showed significantly elevated levels of anti-inflammatory cytokines compared to the DOX group,and the levels of NLRP3,caspase-1,and IL-1βin cells and tissues were reduced by the cleavage of translated proteins.It was concluded that CPA demonstrated an ability to mitigate the impact of DOX-induced cardiac toxicity in both in vivo and in vitro experiments.
