复方蛹虫草口服液镇静催眠作用及安全性

摘要 目的探讨复方蛹虫草口服液(Compound Cordyceps militaris oral liquid,CCMOL)的镇静催眠作用及安全性。方法通过小鼠自主活动试验、戊巴比妥钠协同试验观察CCMOL对小鼠的镇静作用及对对氯苯丙氨酸致失眠小鼠的催眠作用。采用小鼠急性毒性试验、遗传毒性试验评价CCMOL的安全性。结果与失眠模型小鼠比较,3、15、30 mL/kg体质量的CCMOL均可显著降低小鼠自主活动次数(P<0.05或P<0.01),增加阈下剂量戊巴比妥钠小鼠入睡数量,缩短阈剂量戊巴比妥钠小鼠睡眠潜伏期(P<0.01),延长睡眠时间(P<0.01)。CCMOL对雌、雄小鼠的急性经口最大耐受剂量均大于67 mL/kg。小鼠骨髓微核试验、精子畸形试验及细菌回复突变试验(Ames)结果均为阴性。结论CCMOL具有良好的镇静催眠作用,且未见明显毒副作用。 Objective To explore the sedative-hypnotic activity and safety of compound Cordyceps militaris oral liquid(CCMOL).Method The sedative effect of CCMOL was observed by mouse locomotive activity,and Pentobarbital sodium synergistic experiment was used to study the hypnotic effect of CCMOL on insomnia mice induced by p-chlorophenylalanine(PCPA).The safety of CCMOL was evaluated by acute toxicity test and genetic toxicity test in mice.Results Compared to the model group,CCMOL with doses of 3,15,30 mL/kg BW decreased the number of spontaneous activity in mice(P<0.05,P<0.01),increased the number of sleeping mice,shortened the sleep latency of mice(P<0.01)and prolonged the sleep time of mice induced with Pentobarbital sodium at a threshold dose(P<0.01).The oral maximum tolerated dose of CCMOL was more than 67 mL/kg.The results of mice bone marrow micro-nuclear test,sperm malformation test and Ames test were negative.Conclusion CCMOL has effective sedative-hypnotic activity on mice,and has no significant toxic effect.
