摘要
目的探讨地肤子黄酮乙醇提取法的最佳提取条件,并证明提取的地肤子黄酮具有抗炎作用。方法采用正交试验方法探究乙醇提取法提取地肤子黄酮最佳条件。选择SPF级小鼠40只,随机分为空白对照组,地肤子黄酮低、中、高剂量组和阳性药物对照组。用二甲苯涂抹小鼠耳廓使其肿胀,建立小鼠急性炎症模型,并连续灌胃2周。称小鼠耳片质量,计算耳廓肿胀抑制率,同时检测血液中肿瘤坏死因子γ(Tumor necrosis factorγ,TNF-γ)水平。结果地肤子黄酮最佳提取条件为乙醇75%、温度60℃、提取时间4 h、固液比110(m V)、提取次数3次。地肤子黄酮低、中、高剂量组与阳性药物组小鼠耳廓肿胀抑制率、TNF-γ水平与空白对照组比较差异具有统计学意义(P<0.01),地肤子黄酮低、中、高剂量组与阳性药物组比较差异无统计学意义(P>0.05)。结论地肤子黄酮高剂量组与非甾体类药物阿司匹林抗炎效果相似,可作为更安全有效的治疗皮炎替代药物。
Objective To investigate the optimal extraction condition of flavonoids of Fructus kochiae,and to prove that flavonoids of Fructus kochiae has anti-inflammatory effect.Method The best condition for extracting flavonoids of Fructus kochiae by alcohol extraction method were explored,and the mice were divided into control groups,the low,medium and high dose groups of flavonoids of Fructus kochiae and the nonsteroidal drug group,xylene was applied to the auricle of mice to swelling,and a mouse acute inflammation model was established,the inhibition rate of auricular swelling and tumor necrosis factorγ(TNF-γ)were measured.Results The optimal extraction conditions for flavonoids of Fructus kochiae were ethanol 75%,temperature 60℃,extraction time 4 h,material ratio 110(m V),and extraction times 3.The inhibition rate of auricular swelling in mice with low,medium and high dose group and nonsteroidal drug group of flavonoids of Fructus kochiae compared with the blank control group,the difference was statistically significant(P<0.01),and compared with the positive drug group,there was no significant difference(P>0.05).The results showed that flavonoids of Fructus kochiae had anti-inflammatory effect.Conclusion The results demonstrated that flavonoids of Fructus kochiae in the high dose group had similar anti-inflammatory effects to the nonsteroidal drug aspirin,and could be used as an safer and effective alternative medicine to the treatment of dermatitis.
