不同提取方法对三七花多糖生物活性的影响

Effects of Different Extraction Methods on Bioactivity of Polysaccharide from Flower Bud of Panax notoginseng

摘要 以多糖含量、抗氧化性、益生活性、体外降糖降脂能力为指标,比较传统回流法、酶法结合回流提取法和酶法辅助超声法3种提取方法对三七花多糖生物活性的影响,并用最优方法提取的三七花多糖通过秀丽隐杆线虫模型来验证其是否有延缓衰老的活性。结果表明:酶解辅助超声法提取的三七花多糖含量最高,可达146.07 mg/g,体外抗氧化能力(1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基、2,2-联氨-双-3-乙基苯并噻唑啉-6-磺酸(2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基和羟自由基清除率、总还原力和铁离子还原能力(Ferric ion reducing antioxidant power,FRAP)值)最强,并且对植物乳杆菌与干酪乳杆菌的增殖作用益生效果最佳。对三七花多糖降糖降脂能力进行综合评价,传统回流法对三七花多糖降糖降脂能力(α-葡萄糖苷酶、α-淀粉酶抑制率和胰脂肪酶抑制率)影响最大。综合考虑,选择酶法辅助超声法提取的三七花多糖进行秀丽隐杆线虫实验,与对照组相比,0.125 mg/mL的三七花多糖延长线虫寿命8.34%。该研究为三七花多糖的不同提取方法对生物活性的影响提供了理论依据。 The bioactivities of Panax notoginseng flower bud polysaccharide which was extracted by reflux,enzyme-reflux extraction and enzyme-assisted ultrasound extraction were compared with polysaccharide content,antioxidant activity,probiotic activity,in vitro hypoglycemia and lipid-lowering as indicators.The polysaccharide extracted from Panax notoginseng flower bud was tested by means of C.elegans model to verify its anti-aging activity.The results showed that the polysaccharide content extracted by enzymolysis assisted ultrasonication method was the highest,reaching 146.07 mg/g,and the antioxidant capacity(1,1-diphenyl-2-picrylhydrazyl(DPPH)radical and 2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation and hydroxyl radical scavenging activities,total reducing power and ferric ion reducing antioxidant power(FRAP))was the strongest,and the probiotic effect on the proliferation of L.plantarum and L.casei was the best.The hypoglycemic and lipid-lowering ability of Panax notoginseng flower bud polysaccharide was evaluated comprehensively.Reflux method had the greatest effect on the hypoglycemic and lipid-lowering ability of polysaccharide(α-glucosidase,α-amylase and pancreatic lipase inhibition rate).After comprehensive consideration,the polysaccharide by enzyme-assisted ultrasonography was selected for C.elegans experiment.Compared with the control group,0.125 mg/mL polysaccharide extended the life of C.elegans by 8.34%.This study provided theoretical basis for the effects of different extraction methods on biological activities of Panax notoginseng flower bud polysaccharide.
