摘要
目的:探讨Brahma相关基因1相关因子57(BAF57)在结直肠癌(CRC)组织中的表达及其对患者预后的影响。方法:选择2016年1月—2018年1月河北省衡水市人民医院收治的110例CRC患者的癌组织和癌旁组织作为研究对象,分别采用荧光定量PCR(qPCR)和Western blot方法检测癌及癌旁组织中BAF57的mRNA和蛋白表达,分析CRC癌组织中BAF57表达水平与临床病理指标的关系;采用受试者工作特征(ROC)曲线分析BAF57表达水平诊断CRC的价值,以癌组织中BAF57表达水平均值为基准,将患者分为高低表达组。采用Kaplan-Meier和Log-rank检验进行BAF57高、低表达水平组间的生存分析。结果:CRC癌组织中BAF57 mRNA和蛋白的表达水平均显著高于癌旁组织(P<0.05),且其表达水平与患者的TNM分期均呈正相关(r分别为0.913和0.925,均为P<0.01),与癌组织分化程度呈负相关(r分别为-0.781和-0.888,均为P<0.01);与淋巴结转移呈正相关性(r分别为0.744和0.743,均为P<0.01)。ROC分析显示,BAF57 mRNA和蛋白诊断CRC的曲线下面积分别为0.976和0.915。生存分析表明,BAF57 mRNA和蛋白高表达组的中位生存时间均明显短于低表达组,差异均有统计学意义(均为P<0.05)。结论:BAF57在CRC组织中高表达,且与患者恶性进展及不良预后呈正相关,BAF57有望成为CRC患者预后不良的预测指标。
OBJECTIVE:To investigate the expression of Brahma-related gene 1-associated factor 57(BAF57)in colorectal cancer(CRC)tissues and its impact on prognosis of patients.METHODS:The cancer and adjacent tissues of 110 CRC patients admitted to our hospital from January 2016 to January 2018 were selected as the study subjects.BAF57 expression was detected by Western blot and qPCR,respectively.Relationships between BAF57 expression levels and pathological parameters in the tissues were analyzed.Receiver operating characteristic curve and subject operator(ROC)curve were used to determine the relationships between BAF57 expression and diagnosis value.Survival time was analyzed by Kaplan-Meier and Log-rank.RESULTS:The BAF57 mRNA and protein levels were significantly higher in the CRC than those in the adjacent tissues(P<0.05).Both levels were positively correlated with TNM stages(r=0.913,P<0.01;r=0.925,P<0.01),negatively related with tissue differentiation(r=-0.781,P<0.01;r=-0.888,P<0.01);and positively with lymph node metastasis(r=0.744,P<0.01;r=0.743,P<0.01).ROC analysis showed that the area under the curve for BAF57 mRNA and protein for CRC diagnosis was 0.976 and 0.915,respectively.Survival analysis indicated that the median survival time of the BAF57 mRNA high-expression group(48.5 month)was significantly lower than the low-expression group(60.5 month,Log-rankχ2=4.985,P<0.05),which was also found between the protein high-and low-expression groups(46.5 and 60.0 months,respectively;Log-rankχ2=4.469,P<0.05).CONCLUSION:BAF57 was highly expressed in CRC tissues and was positively associated with malignant progression and poor prognosis in patients.
