摘要
目的观察成纤维细胞生长因子2(FGF2)对体外培养胚龄12.5 d(E12.5 d)胎鼠肾组织发育的影响,从而探求它与肾小体发育的关系。方法E12.5 d的胎鼠肾组织分别用0,25,100,200,400 ng/mL FGF2体外培养,培养2,4,6 d后HE染色观察肾组织中肾小体的发育情况;免疫组织化学染色结合体视学对不同浓度FGF2体外培养肾组织中各期肾小体成纤维细胞生长因子受体1(FGFR1)的表达进行半定量分析。结果E12.5 d胎鼠肾组织在体外培养过程中有类似在体的发育过程,经FGF2培养后,可见发育不同阶段的肾小体增多。免疫组织化学染色和体视学结果显示经与0 ng/mL FGF2比较,25,100,200,400 ng/mL FGF2体外培养后,肾组织中FGFR1的表达显著升高(P<0.05),但4个浓度间差异无统计学意义。结论推测在肾脏的发育过程中,FGF2主要通过与FGFR1结合从而促进间充质细胞的聚集和肾小体的发育。
Objective To investigate the effect of the fibroblast growth factor 2(FGF2)on the development of in vitro cultured kidney tissues from the 12.5-day embryonic mice,and explore its relationship with the development of renal corpuscle.Methods The kidney tissues of 12.5-day embryonic mice were in vitro cultured with 0,25,100,200,400 ng/mL FGF2 for 2,4,6 d,and then the development of renal corpuscles was observed by HE staining.Immunohistochemical staining combined with stereo-logical methods were used to observe and semi-quantitatively analyze the expression of FGFR1 in renal corpuscles at different developmental stages.Results The development process of kidney tissues in vitro was similar to the development process in vivo.After cultured with FGF2,the number of renal corpuscles at different stages of development process increased.Immunohistochemical staining and stereo-logical results showed the expression of FGFR1 in renal tissue in vitro cultured with 25,100,200,400 ng/mL FGF2 was significantly higher than that of 0 ng/mL FGF2(P<0.05),but there was no statistically significant difference among four concentration groups.Conclusion FGF2 may promote the aggregation of mesenchymal cells and the development of renal corpuscles by binding to FGFR1 during the kidney development process.
