清肺止咳散HPLC指纹图谱和多成分含量的测定

Determination of HPLC Fingerprint and Multi-ComponentContent of Qingfei Zhike San

为了建立清肺止咳散高效液相色谱(HPLC)指纹图谱和多成分含量测定的方法,为其质量评价提供参考。本试验采用HPLC法,建立12批清肺止咳散样品的HPLC指纹图谱,以《中药色谱指纹图谱相似度评价系统》(2012版)评价指纹图谱的相似度,并结合主成分分析(PCA)和偏最小二乘法判别分析(PLS-DA)筛选质量标志物,同时测定清肺止咳散中被指认的质量标志物的含量。结果显示,12批样品共确定33个共有峰,指认了绿原酸、苦杏仁苷、柚皮苷、黄芩苷、黄芩素、汉黄芩素和白花前胡甲素7个特征性成分;12批样品按不同厂家聚为3个组,筛选出绿原酸、苦杏仁苷、柚皮苷和黄芩苷4个质量标志物,各成分呈良好的线性关系(r>0.999),平均加样回收率为95.2%~101.2%;12批样品中绿原酸、苦杏仁苷、柚皮苷和黄芩苷含量范围分别为0.65~7.09、3.45~8.98、0.95~10.60和4.03~25.83 mg/g。本试验所建立的HPLC指纹图谱和多成分含量测定方法简便、稳定,可为清肺止咳散的质量控制和品质评价提供依据。

In order to establish a method for the high performance liquid chromatography(HPLC)fingerprint and multi-component content determination of Qingfei Zhike San and provide a reference for its quality evaluation,this study utilized HPLC to develop a fingerprint profile of 12 batches of Qingfei Zhike San samples.The similarity of the fingerprints was evaluated using the"Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System"(2012 version),and quality markers were screened through principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA).The content of the identified quality markers in Qingfei Zhike San was also determined.The results showed that 33 common peaks were identified across the 12 batches,and 7 characteristic components were identified,including chlorogenic acid,amygdalin,naringin,baicalin,baicalein,wogonin,and praeruptorin A.The 12 batches of samples were classified into three groups based on different manufacturers,with chlorogenic acid,amygdalin,naringin,and baicalin selected as quality markers.Each component exhibited good linearity(r>0.999),with average recovery rates ranging from 95.2%to 101.2%.The content ranges of chlorogenic acid,amygdalin,naringin,and baicalin in the 12 batches were 0.65-7.09,3.45-8.98,0.95-10.60,and 4.03-25.83 mg/g,respectively.The HPLC fingerprint and multi-component determination method established in this study is simple and stable,providing a basis for the quality control and evaluation of Qingfei Zhike San.
