UPLC-QDa法同时测定老鹰茶提取物中8种黄酮类化合物

摘要 建立超高效液相色谱-四级杆质谱检测器(UPLC-QDa)法同时测定老鹰茶提取物中8种黄酮类成分的快速高效分析方法。色谱柱Waters ACQUITY UPLC BEH C_(18)(50 mm×2.1 mm,1.7μm),以水(含0.1%甲酸)-乙腈(含0.1%甲酸)为流动相进行梯度洗脱,流速:0.4 mL/min,柱温35℃,电喷雾离子源负离子(ESI-)模式扫描。8种黄酮化合物的峰基线完全分离,线性关系良好,相关性系数均大于0.996,检出限为0.018~0.294μg/mL,定量限为0.110~0.294μg/mL。仪器精密度和重复性良好,RSD(n=6)分别为0.72%~1.25%、1.79%~3.96%。平均回收率在94.1%~108.6%之间,RSD为2.1%~4.4%。本方法快速高效、灵敏度高、重复性好、操作简单,可用于老鹰茶提取物中黄酮类化合物的含量测定,为后续其质量标准的提升提供参考。 A rapid and efficient analytical method for the simultaneous determination of 8 flavonoid components in hawk tea extract using ultra high performance liquid chromatography-quadrupole detector(UPLC-QDa)was established.The chromatographic column used Waters ACQUITY UPLC BEH C_(18)(50 mm×2.1 mm,1.7μm),with a mobile phase of water(containing 0.1%formic acid)and acetonitrile(containing 0.1%formic acid)in gradient elution,flow rate was 0.4 mL/min,column temperature was 35℃,and scanning in negative ion(ESI-)mode of electrospray ionization source.The 8 flavonoid compounds were completely separated with excellent baseline resolution,good linearity with correlation coefficients was all above 0.996,detection limits ranged from 0.018 to 0.294μg/mL,and quantitation limits were from 0.110 to 0.294μg/mL.The instrument precision and repeatability were good,with RSD(n=6)of 0.72%to 1.25%and 1.79%to 3.96%,respectively.The average recovery rate ranged from 94.1%to 108.6%,with RSD from^(2).1%to 4.4%.This method is rapid,highly efficient,sensitive,repeatable,and simple to operate,suitable for the determination of flavonoid content in hawk tea extracts,providing a reference for subsequent enhancement of its quality standards.
