摘要
背景变应性鼻炎是常见的变态反应性疾病,临床症状轻重程度与血清IgE含量关系密切。IL-10家族细胞因子抑制过度炎症反应、上调先天免疫和促进组织修复,在发病机制中起关键作用。中医益气宣化法治疗变应性鼻炎临床疗效佳。目的以OVA诱导变应性鼻炎动物模型为研究对象,阐释益气宣化方影响鼻黏膜IL-10分泌从而抑制变应性鼻炎的机制。方法24只小鼠随机分为空白对照组6只和变应性鼻炎模型组18只。建立小鼠变应性鼻炎模型。模型组分为三组,分别予生理盐水,益气宣化汤,氯雷他定干预。观察各组小鼠行为学评分。通过ELISA分析检测各组小鼠血清IgE含量及鼻黏膜IL-10含量。结果益气宣化汤能够有效降低变应性鼻炎小鼠的临床表征(P<0.05),有效降低变应性鼻炎小鼠外周血中IgE含量(P<0.05),有效升高鼻黏膜组织IL-10的含量(P<0.05)。结论益气宣化方通过调节血清IgE含量和鼻黏膜IL-10分泌,达到治疗作用。
Background Allergic rhinitis is a common allergic disease,the severity of clinical symptoms is closely related to the level of IgE in serum.The IL-10 family of cytokines plays a crucial role in the pathogenesis of allergic rhinitis by inhibiting excessive inflammatory response,upregulating innate immunity,and promoting tissue repair mechanisms.The traditional Chinese medicine Yiqixuanhua Formula has a good clinical effect on treating allergic rhinitis.Objective Taking OVA induced allergic rhinitis animal model as the research object,this study elucidates the mechanism of Yiqixuanhua Formula affecting the secretion of IL-10 in nasal mucosa and inhibiting allergic rhinitis.Methods A total of 24 mice were randomly divided into a blank control group of 6 and an allergic rhinitis model group of 18.Mouse model of allergic rhinitis was established.The model group was divided into three groups,each receiving physiological saline,Yiqixuanhua Formula and loratadine intervention.The behavioral scores of mice were observed.Serum IgE content and nasal mucosal IL-10 content of mice were detected through ELISA analysis.Result Yiqixuanhua Formula can effectively reduce the clinical manifestations of allergic rhinitis mice(P<0.05),reduce the IgE content in the peripheral blood of allergic rhinitis mice(P<0.05),and increase the IL-10 content in nasal mucosa tissue(P<0.05).Conclusion Yiqixuanhua Formula has the effect of treating allergic rhinitis by regulating serum IgE content and nasal mucosal IL-10 secretion.
