摘要
本研究针对传统的15 d传代方法,改用7 d石蜡保存法和甘油肉汤保存法,探索适合莱士曼氏乳酸杆菌传代培养方法。通过长期实验验证,分析标准曲线和相对标准偏差(Relative Standard Deviation,RSD),得到这两种方法标准曲线线性关系r^(2)均> 0.99,相对标准偏差RSD分别为2.5%~3.7%和2.9%~7.2%。证实了用7 d石蜡保存法、甘油肉汤保存法这两种方法保存的莱士曼氏乳酸杆菌活性稳定,活化周期短,可满足维生素B_(12)检测要求。
In this study,the traditional method of 15-day passage method was changed to 7-day passage method with liquid paraffin and glycerin broth preservation method to explore a suitable subculture method of Lactobacillus leishmanni.The standard curve and the relative standard deviation(RSD)of these two methods were analyzed through long-term experimental verification.The results showed that these two methods standard curve linear relationship r^(2) was>0.99,and the RSD were 2.5%-3.7%and 2.9%-7.2%,respectively.This study proved that Lactobacillus leishmanni saved by the 7-day passage method with liquid paraffin and the glycerin broth preservation method had stable activity and short activation period,which can meet the requirement of vitamin B_(12) detection.
