摘要
运用网络药理学的研究方法,深入探索菝葜治疗寻常型银屑病的作用机制。利用中药系统药理学数据库筛选出菝葜的活性成分及其作用靶点,并与寻常型银屑病的靶点取交集,得到菝葜治疗寻常型银屑病的潜在作用靶点。针对这些靶点,构建了PPI网络,并进一步进行GO富集分析和KEGG通路富集分析。分析结果显示:菝葜含有13个活性成分与76个靶点蛋白相互作用,共同参与了25条与寻常型银屑病密切相关的关键通路。广泛的生物过程被生物学功能所涵盖,包括调节脂质代谢过程、大分子复合物、酶结合等。菝葜治疗银屑病的主要富集通路为IL-17信号通路和TNF信号通路。分子对接实验进一步证实,菝葜的核心成分能够与核心靶点有效结合,从而发挥治疗作用。本研究初步揭示了菝葜作为治疗寻常型银屑病的有效中药,其疗效可能与对IL-17和TNF信号通路等关键靶点的调节有关。该结果为菝葜在银屑病治疗中的应用提供更为深入的理论依据。
The action mechanism of BaQia in the treatment of psoriasis vulgaris was explored in depth by us⁃ing the research method of network pharmacology.The active ingredients of BaQia and their targets were screened using the traditional Chinese medicine(TCM)systemic pharmacology database,and intersected with the targets of psoriasis vulgaris,thus obtained the potential targets of BaQia for treatment of psoriasis vulgaris.A protein PPI net⁃work was constructed for these targets,and further GO function analysis and KEGG pathway enrichment analysis were performed.The analysis results showed that BaQia contained 13 active components,which interacted with 76 target proteins and collectively participated in 25 key pathways closely related to psoriasis vulgaris.These biological functions covered a wide range of biological processes,including regulation of lipid metabolic processes,macromolec⁃ular complexes,enzyme binding,etc.The main enriched pathways for BaQia treatment of psoriasis were IL-17 sig⁃nalling pathway and TNF signalling pathway.Molecular docking experiments further confirmed that the core compo⁃nents of BaQia could effectively bind to these core targets to exert therapeutic effects.The study preliminarily re⁃vealed that the efficacy of BaQia as an effective traditional Chinese medicine in the treatment of psoriasis vulgaris might be related to the modulation of key targets such as IL-17 and TNF signalling pathways.This result provided a more in-depth theoretical basis for the application of BaQia in the treatment of psoriasis.
