摘要
目的探究青年CD-1雄性小鼠经历盲肠结扎穿孔(CLP)术后发生认知障碍及其恢复情况。方法取7~8周龄CD-1雄性小鼠若干分别造模,设立CLP组和假手术(Sham)组。造模成功后,分别将存活的Sham和CLP小鼠喂养至术后第10、10、20、30和60天进行六臂放射状水迷宫(RAWM)和物体位置再认测试。结果与Sham组相比,CLP小鼠术后30 d以内在RAWM中的学习期错误次数和潜伏期均高于Sham组(P<0.05),而在术后60 d差异无统计学意义(P>0.05);CLP小鼠术后60 d以内的RAWM记忆期错误次数和潜伏期都显著高于Sham组(P<0.05);在物体位置再认实验中,CLP小鼠术后10 d和20 d的10 min延迟、24 h延迟的新位置优先指数均低于Sham组(P<0.05),而术后30 d和60 d的10 min延迟、24 h延迟的新位置优先指数与Sham组比较差异均无统计学意义(P>0.05)。结论青年期CD-1雄性小鼠在CLP术后60 d时仍有显著性空间记忆损害,且RAWM是较敏感的评估方法。
Objective To investigate the cognitive impairment and its recovery in young male CD-1 mice after cecal ligation and perforation(CLP).Methods A number of CD-1 male mice aged 7~8 weeks were modeled respectively,and CLP group and sham operation group(Sham group)were set up.After successful modeling,the surviving Sham and CLP mice were fed until the 10th,10th,20th,30th and 60th day after surgery for radial six-arm water maze(RAWM)and object position re-recognition,respectively.Results Compared with Sham group,the number of learning errors and latency of RAWM in CLP mice within 30 days after surgery were higher than those in Sham group(P<0.05),but there was no significant difference at 60 days after surgery(P>0.05).The num-ber of RAWM memory errors and latency of CLP mice within 60 days after operation were significantly higher than those of Sham group(P<0.05).In the object position re-recognition experiment,the new position preference index of 10 min delay and 24 h delay of CLP mice were lower than those of Sham group at 10 and 20 days after op-eration(P<0.05).There were no significant differences in the new position preference index of 10 min delay and 24 h delay at 30 and 60 days after surgery compared with Sham group(P>0.05).Conclusion Young CD-1 male mice still had significant spatial memory im-pairment at 60 days after CLP surgery,and RAWM was a more sensi-tive evaluation method.
