测定血清中21种元素的前处理方法研究

摘要 目的采用电感耦合等离子体质谱技术,测定血清中21种元素含量,重点优化血清中多元素测定的样品前处理方法。方法采用1%硝酸,1%TtitonX-100,0.01%吡咯二硫代氨基甲酸铵与1%乙醇混合溶剂稀释血清样品20倍后,进入ICP-MS仪器检测Mn、Co、Ni、Cd、La、Ce、W、Hg、Tl、Mo、Sb、Fe、Cu、Zn、Se、Al、As、Sr、Pb、Sn和Cr 21种元素的含量。结果血清中21种元素在线性范围内线性良好(r>0.995);检出限为0.14~15.90μg/L。方法用于孕妇血清样本检测,低高浓度2个水平的血清加标回收率为82.2%~121.5%,相对标准偏差小于10%。采集639名孕妇血清样本进行检测,21种元素中除Tl、W、La、Ce、Hg,其余元素均有检出。结论通过条件优化,采用混合溶剂稀释法联合ICP-MS对血清中21种金属元素分析,方法快速、简便、准确,适宜用于评估人体元素暴露水平。 Objective The contents of 21 elements in serum was determined by inductively coupled plasma mass spectrometry,and the sample pre-treatment method for multi-element determination in serum was optimized.Methods The serum samples were diluted 20 times with 1%nitric acid,1%TItonX-100,0.01%ammonium pyrrole dithiocarbamate and 1%ethanol.The content of 21 elements such as Mn,Co,Ni,Cd,La,Ce,W,Hg,Tl,Mo,Sb,Fe,Cu,Zn,Se,Al,As,Sr,Pb,Sn and Cr was detected by the ICP-MS instrument.Results The 21 elements in serum had good linearity in the linear range(r>0.995),and the detection limits ranged from 0.14 to 15.90μg/L.The method was used for the detection of serum samples from pregnant women.The recovery rates of serum spiked at two levels of low and high concentrations were within 82.2.0%-121.5%,with a relative standard deviation of less than 10%.The serum samples of 639 pregnant women were collected for detection.Among the 21 elements,except for Tl,W,La,Ce,and Hg,all other elements were detected.Conclusions By optimizing the conditions,the mixed solvent dilution method combined with ICP-MS was used to analyze 21 metal elements in serum.The method is fast,simple,and accurate,it provids data reference and technical support for evaluating human element exposure levels.
