七宝美髯酊治疗脱发的皮肤药效学研究

Cutaneous pharmacodynamics of Qibao Meiran tincture in treatment of alopecia

目的:研究七宝美髯酊对SD大鼠毛发、毛囊生长周期的影响。方法:将60只SD大鼠随机分为6组:10%、30%、50%、70%七宝美髯酊组,米诺地尔组,空白对照组,每组各10只。采用外用脱毛膏的方式进行造模,面积约40 cm^(2)(8 cm×5 cm):造模次日进行药物干预,七宝美髯酊各剂量组予不同浓度七宝美髯酊外擦,米诺地尔组外擦5%米诺地尔酊,均在用药后局部均匀按摩3~5圈至药物吸收,1 mL/次,2次/d,连续干预21 d,空白对照组不予药物干预。对大鼠皮肤颜色状况进行观察、记录。干预结束后,经随机方式选择12处新生毛发,借助百分电子数显游标卡尺实施测定并记录;采集动物观察区的一块皮肤,进行苏木精-伊红(HE)染色,用显微镜查看大鼠真皮层厚度、毛囊数目与形态学变化。结果:与空白对照组比较,七宝美髯酊各剂量组大鼠皮肤由粉红色到白色的变化时间均缩短,毛发长度增长及毛囊数量增加,其中以50%七宝美髯酊剂量组时间最短,差异均有统计学意义(P<0.05)。干预第21天,50%七宝美髯酊组的毛发长度及毛囊数量与米诺地尔组相当(P>0.05),且优于其他各组(P<0.05)。30%、50%、70%七宝美髯酊组皮肤真皮层厚度较空白对照组增加(P<0.01),且与米诺地尔组差异无统计学意义(P>0.05)。结论:七宝美髯酊可加快大鼠毛发生长、促进大鼠毛囊生长期转化进程,其中50%浓度疗效与米诺地尔相当。

Objective:To investigate the effect of Qibao Meiran tincture on the cyclic growth of hair and hair follicles in Sprague-Dawley(SD) rats.Methods:A total of 60 SD rats were randomly divided into 10%,30%,50%,and 70% Qibao Meiran tincture groups,minoxidil group,and blank control group,with 10 rats in each group.External application of hair removal cream was performed for modeling with an area of about 40 cm~2(8 cm × 5 cm),and drug intervention was performed since the day after modeling.The rats in the Qibao Meiran tincture groups were given external application of Qibao Meiran tincture at different concentrations,while those in the minoxidil group were given external application of 5% minoxidil tincture,at a volume of 1 mL each time,twice a day for 21 consecutive days,and the drugs were rubbed to absorption;the rats in the blank control group were not given any drug intervention.The color and condition of the skin were observed and recorded.After intervention,12 sites of newly grown hair were randomly selected and were measured and recorded using a vernier caliper with percentile electronic digital display;a piece of skin was collected from the observation area,and HE staining was performed to observe the thickness of the corium layer,the number of hair follicles,and morphological changes.Results:Compared with the blank control group,the Qibao Meiran tincture groups had a significant reduction in the time for the skin to change from pink to white color and significant increases in the length of hair and the number of hair follicles,and the 50% Qibao Meiran tincture group showed the shortest time(P<0.05).On day 21 of intervention,the 50% Qibao Meiran tincture group was comparable to the minoxidil group in terms of the length of hair and the number of hair follicles(P>0.05),with significantly better results than the other groups(P<0.05).Compared with the blank control group,the 30%,50%,and 70% Qibao Meiran tincture groups had a significant increase in the thickness of the corium layer(P<0.01),with no significant difference compared with the minoxidil group(P>0.05).Conclusion:Qibao Meiran tincture can accelerate hair growth in rats and promote transformation during the growing stage of hair follicles,and 50% Qibao Meiran tincture has a comparable clinical effect to minoxidil.
