摘要
目的探讨瞬时受体电位M7通道(TRPM7)和B细胞迁移基因2(BTG2)在宫颈癌组织中的表达及临床意义。方法选取2018年5月至2020年5月石家庄市妇幼保健院收治的110例宫颈癌患者作为研究对象,术中取其肿瘤组织及癌旁组织,根据随访情况分为预后良好组与预后不良组。采用免疫组化法检测癌旁组织和肿瘤组织中TRPM7、BTG2蛋白表达,分析TRPM7、BTG2蛋白表达水平与患者临床病理特征的关系,比较预后不良组与预后良好组肿瘤组织中TRPM7、BTG2蛋白表达,采用Cox回归分析宫颈癌患者预后的影响因素,采用Kaplan-Meier曲线分析TRPM7、BTG2水平与宫颈癌患者预后的关系。结果与癌旁组织比较,肿瘤组织中TRPM7蛋白表达水平升高,BTG2蛋白表达水平降低(P<0.05)。TRPM7、BTG2蛋白表达水平与肿瘤分化程度、淋巴结转移、恶性肿瘤国际临床病理(TNM)分期有关(P<0.05)。与预后良好组比较,预后不良组肿瘤组织中TRPM7蛋白表达水平升高,BTG2蛋白表达水平降低(P<0.05)。TRPM7、TNM分期(Ⅲ期)、肿瘤低分化程度、淋巴结转移是宫颈癌患者预后的危险因素(P<0.05),BTG2是宫颈癌患者预后的保护因素(P<0.05)。TRPM7高表达组3年生存率低于TRPM7低表达组(P<0.05);BTG2高表达组3年生存率高于BTG2低表达组(P<0.05)。结论宫颈癌患者肿瘤组织中TRPM7蛋白表达上调,BTG2蛋白表达下调,均与宫颈癌预后有关。
Objective To investigate the expression and clinical significance of transient receptor potential melastatin 7(TRPM7)and B cell translocation gene 2(BTG2)in cervical cancer tissue.Methods A total of 110 cervical cancer patients admitted to Shijiazhuang Maternal and Child Health Hospital from May 2018 to May 2020 were selected as the research objects,and their tumor and adjacent tissues were taken during surgery.They were grouped into a good prognosis group and a poor prognosis group according to the follow-up condition.Immunohistochemical method was applied to detect the expression of TRPM7 and BTG2 proteins in adjacent and tumor tissues.The relationship between TRPM7 and BTG2 protein expression levels and the clinicopathological features of patients was analyzed.TRPM7 and BTG2 protein expression in tumor tissues of the poor prognosis group and the good prognosis group were compared.Cox regression was applied to analyze the influencing factors for the prognosis of cervical cancer patients.Kaplan-Meier curve was used to analyze the relationship between TRPM7 and BTG2 levels and the prognosis of cervical cancer patients.Results Compared with adjacent tissues,the expression level of TRPM7 protein in cervical cancer tumor tissue increased and the expression level of BTG2 protein reduced(P<0.05).The expression levels of TRPM7 and BTG2 proteins were related to tumor differentiation,lymph node metastasis,and malignant tumors international clinicopathology tumor node metastasis(TNM)classification(P<0.05).Compared with the good prognosis group,the expression level of TRPM7 protein in the tumor tissue of poor prognosis group increased,while the expression level of BTG2 protein decreased(P<0.05).TRPM7,TNM stage(stageⅢ),low differentiation of tumor and lymph node metastasis were risk factors for the prognosis of cervical cancer patients(P<0.05),and BTG2 is a protective factor for the prognosis of patients with cervical cancer(P<0.05).The 3-year survival rate of patients in the high TRPM7 expression group was lower than that in the low TRPM7 expression group(P<0.05).The 3-year survival rate of patients in the high BTG2 expression group was higher than that in the low BTG2 expression group(P<0.05).Conclusions TRPM7 protein expression is up-regulated and BTG2 protein expression is down-regulated in the tumor tissues of cervical cancer patients,and they are both related to the prognosis of cervical cancer patients.
