瑶医神火灸通过调控Th17/Treg轴免疫功能失衡改善缺血再灌注大鼠模型的作用研究

摘要 目的:探讨瑶医神火灸对缺血再灌注大鼠Th17/Treg免疫平衡的影响。方法:将40只健康SD雄性大鼠随机分为假手术组、模型组、神火灸组及艾灸组4组,使用线栓术制作缺血再灌注模型,在治疗前、治疗14 d后对大鼠神经功能缺损进行评分,使用TTC染色观察不同治疗组脑梗死体积变化,使用RT-PCR法检测各组大鼠脑组织中IL-17 mRNA、IL-6 mRNA、TGF-β2mRNA、IL-10 mRNA的表达水平,并比较Th17/Treg轴比值变化情况。结果:经治疗,神火灸组及艾灸组脑梗死体积与模型组相比减小(P<0.05),同时神火灸组脑梗死体积比艾灸组更小(P<0.05);神火灸组及艾灸组神经功能均改善,其中神火灸组神经功能改善更显著;与模型组及艾灸组相比,瑶医神火灸组IL-17 mRNA、IL-6 mRNA等的表达减少(P<0.05),而TGF-β2 mRNA、IL-10 mRNA水平的表达增加(P<0.05),经过两者比较发现神火灸组IL-6/IL-10比值降低,差异具有统计学意义(P<0.05),提示经过瑶医神火灸干预后Th17/Treg免疫平衡下降,具有统计学意义。结论:瑶医神火灸可以通过降低Th17分泌的促炎因子及升高Treg分泌的抗炎因子的水平,从而调节Th17/Treg免疫平衡轴,减轻脑缺血再灌注损伤。 Objective To investigate the effect of Shenhuo moxibustion of yao medicine ischemia-reperfusion in rats of Th17/Treg immune balance.Methods Forty healthy male SD rats were randomly divided into 4 groups:sham operation group,model group,shenhuo moxibustion group and moxibustion group.Thread block method was used to establish the cerebral ischemia reperfusion injury model in rats.After the reperfusion for 14 days,efforts were made to give neurological score;The expression levels of IL-17 mRNA,IL-6 mRNA,TGF-β2 mRNA and IL-10 mRNA in the brain tissues of rats in each group were detected by RT-PCR,and the changes of Th17/Treg axis ratio were compared.Results After treatment,the nerve function of Shenhuo moxibustion group and moxibustion group were improved,and the nerve function of Shenhuo moxibustion group was improved more significantly;IL-17 mRNA,IL-6 mRNA,TGF-β2mRNA and IL-10 mRNA were detected by RT-PCR.Compared with the model group and moxibustion group,the expressions of IL-17 mRNA and IL-6 mRNA in yaoyishen moxibustion group were decreased(P<0.05).After comparison between the two groups,it was found that the IL-6/IL-10 ratio of shenhuo moxibustion group decreased,and the difference was statistically significant(P<0.05),suggesting that the Immune balance of Th17/Treg decreased after yao Medico moxibustion intervention,which was statistically significant.Conclusion shenhuo moxibustion may reduce the pro-inflammatory factors secreted by Th17 and increase the expression of anti-inflammatory factors secreted by Treg,and regulate the immune balance axis of Th17/Treg.
