摘要
目的:探究增强子RNA(eRNA)ENSR00000313345在头颈部鳞状细胞癌(HNSCC)中的作用。方法:通过Spearman相关性分析检测生存相关eRNA ENSR00000313345与其假定靶基因WWC2之间的共表达。通过实时荧光定量PCR检测HNSCC细胞中eRNA ENSR00000313345的表达;在HNSCC细胞中转染小干扰RNA以敲低eRNA ENSR00000313345,采用CCK⁃8、划痕实验、Transwell迁移实验探讨eRNA ENSR00000313345对HNSCC细胞增殖、迁移的影响。在HNSCC细胞中转染小干扰RNA以敲低WWC2,通过CCK⁃8、划痕实验、Transwell迁移实验以检测WWC2对HNSCC细胞增殖、迁移的影响。通过实时荧光定量PCR和Western blot检测eRNA ENSR00000313345与WWC2的相关性。结果:WWC2的表达量与EN⁃SR00000313345呈正相关。eRNA ENSR00000313345在HNSCC细胞系中的表达量高于口腔黏膜角化上皮细胞系(P<0.05)。敲低eRNA ENSR00000313345可以抑制HNSCC细胞的增殖、迁移(P<0.05)。敲低WWC2能够抑制HNSCC细胞的增殖、迁移(P<0.05)。敲低eRNA ENSR00000313345时,WWC2的表达水平随之下调(P<0.05)。结论:eRNA ENSR00000313345可以通过调控WWC2表达进而促进HNSCC细胞的增殖与迁移。
Objective:To identify the role of eRNA ENSR00000313345 in head and neck squamous cell carcinoma(HNSCC).Methods:Co⁃expression between individual survival⁃related eRNA ENSR00000313345 and its putative target gene WWC2 was identi⁃fied by Spearmans correlation analysis.This expression of eRNA ENSR00000313345 in HNSCC cell lines was detected by qRT⁃PCR technology.Small interfering RNA was transfected into HNSCC cell lines to knock down eRNA ENSR00000313345,followed by func⁃tional experiments including CCK⁃8 assay,cell scratch assay,and Transwell migration assay to verify the effect of eRNA EN⁃SR00000313345 on HNSCC.Small interfering RNA was transfected into HNSCC cell lines to knock down WWC2,and CCK⁃8 assay,cell scratch assay,and Transwell migration assay were performed to detect the effect of WWC2 on HNSCC.The correlation between eR⁃NA ENSR00000313345 and WWC2 was examined using qRT⁃PCR and Western blot techniques.Results:The expression level of WWC2 was positively correlated with eRNA ENSR00000313345.eRNA ENSR00000313345 exhibited higher expression levels in HNSCC cell lines compared to oral mucosal keratinocyte cell lines(P<0.05).Knocking down eRNA ENSR00000313345 suppressed the proliferation and migration capabilities of HNSCC cell lines(P<0.05).Knocking down WWC2 inhibited the proliferation and mi⁃gration abilities of HNSCC cell lines(P<0.05).The expression levels of WWC2 decreased concomitantly with the knockdown of eRNA ENSR00000313345(P<0.05).Conclusions:eRNA ENSR00000313345 can promote the proliferation and migration of HNSCC cells by regulating WWC2 expression.
