摘要
目的探讨原发性干燥综合征(pSS)患者外周血hsa_circ_0045800的表达及其在pSS诊断中的价值。方法采用微阵列芯片技术测序筛查4例pSS患者和4例对照者的外周血差异circRNAs;并以RT-qPCR验证50例pSS患者和50例对照者外周血的hsa_circ_0045800的表达差异;通过受试者工作特征(ROC)曲线评估外周血中hsa_circ_0045800表达水平对pSS的诊断价值。结果pSS患者外周血中hsa_circ_0045800表达水平高于对照者,pSS患者治疗后hsa_circ_0045800表达水平低于治疗前(P<0.05);pSS患者hsa_circ_0045800的表达水平与病情评估指数(ESSDAI)、抗核抗体(ANA)滴度、血沉(ESR)均呈正相关(P<0.05);ROC曲线显示:血清hsa_circ_0045800表达水平曲线下面积(AUC)为0.927,敏感度为74%,特异度为94%,对p SS的诊断效能较好。结论hsa_circ_0045800可能与pSS的发生、发展有关,并且有潜力成为pSS诊断、评估病情严重程度及预后的生物标志物。
Objective To investigate the expression of hsa_circ_0045800 in peripheral blood of patients with primary Sjögren’s syndrome(pSS)and evaluate its value in the diagnosis of pSS.Methods The changes of circRNA in peripheral blood of 4 pSS patients and 4 controls were selected for microarray screening.RT-qPCR was used to verify the differences in the expression of hsa_circ_0045800 in peripheral blood of 50 pSS patients and 50 controls.Receiver operating characteristic(ROC)curve was used to evaluate the diagnostic value of hsa_circ_0045800 expression level in peripheral blood for pSS.Results The expression level of hsa_circ_0045800 in peripheral blood of pSS patients was higher than that of healthy control group,and the expression level of hsa_circ_0045800 in pSS patients after treatment was lower than that before treatment(P<0.05);The expression level of hsa_circ_0045800 in pSS patients was positively correlated with disease assessment index(ESSDAI),ANA titer and erythrocyte sedimentation rate(ESR)(P<0.05);ROC curve showed that the expression level of serum hsa_circ_0045800 had a good diagnostic efficacy for pSS,with an area under the curve(AUC)of 0.927,a sensitivity of 74%,and a specificity of 94%.Conclusion Hsa_circ_0045800 may be related to the occurrence and development of pSS,and has the potential to be a biomarker for the diagnosis,evaluation of disease severity and prognosis of pSS.
