摘要
【目的】探究黄花棘豆对小鼠肝脏内质网应激和损伤的影响,为高原动物黄花棘豆中毒机理研究提供理论依据。【方法】通过构建小鼠黄花棘豆病理模型,HE染色检测小鼠肝脏组织形态变化,RT-qPCR检测小鼠肝脏内质网应激基因(Grp78、Chop、Xbp1、Atf4、Atf6)表达变化,WB检测小鼠肝脏内质网应激蛋白(GRP78、CHOP)表达变化,明确黄花棘豆对小鼠肝脏内质网应激和损伤的影响。【结果】与对照组相比,饲喂黄花棘豆小鼠出现病理症状,体重显著下降,肝脏质量降低,肝细胞索紊乱,肝小叶排列松散,肝细胞变形、细胞坏死,肝脏内质网应激相关基因和蛋白表达显著升高。【结论】说明黄花棘豆导致小鼠肝脏发生内质网应激反应和肝脏损伤。
【Objective】This study aimed to study the effects of Oxytropis ochrocephala on endoplasmic re⁃ticulum(ER)stress and damage in mouse liver,thus providing a theoretical basis for the poisoning mechanism of Oxytropis ochrocephala in plateau animals.【Method】In this study,the pathological model of Oxytropis ochrocephala-fed mouse was established.The morphological changes in mouse liver tissue were detected by HE staining.The expressions of ER stress-related genes and proteins(Grp78,Chop,Xbp1,Atf4,Atf6)in mouse liver were detected by RT-qPCR and WB.The experiments were to clarify the effects of Oxytropis ochrocephala on ER stress and damage in mouse liver.【Result】Com⁃pared to control group,the mouse fed Oxytropis ochrocephala showed pathological symptoms,significant weight loss,decreased liver weight,disordered hepatocyte cord,loose arrangement of hepatic lobules,deformation and necrosis in liver cells,and significant elevation in the expressions of ER stress-related genes and proteins.【Conclusion】In conclusion,Oxytropis ochrocephala can induce ER stress response and damage in mouse liver.
