模型化研究YAP与Notch信号间的串扰动力学

Modeling the Crosstalk Dynamics between YAP and Notch Signals

摘要 基于Hill动力学与Michaelis-Menten方程,建立相对完整的理论模型,研究YAP(Yes-Associated Protein)与Notch信号间的串扰动力学.研究发现,YAP激活可诱导Notch信号表达提升,增强的Notch信号会间接导致YAP表达减弱,揭示了YAP与Notch调控信号间的串扰相互作用.基于本文模型结果可以预测,当YAP与Notch信号串扰存在时,YAP会诱发Notch信号异常;同理,Notch信号异常也会致使YAP信号通路受到影响,在正常细胞反应过程中,YAP与Notch信号动态协同调节细胞进程,这一结果可用于预测存在YAP与Notch信号异常激发的病变.本文所得理论结果符合实验观测,进一步深刻揭示了YAP与Notch信号间串扰动力学的物理机制,可为后续进一步研究YAP与Notch信号串扰调控作用提供有价值的参考. Based on Hill dynamics and Michaelis-Menten equation,a relatively complete theoretical model was established to study the crosstalk dynamics betweenYAPand Notch signals.We found thatYAPactivation can induce an increase in Notch expression.The enhanced Notch signal indirectly leads to a decrease in YAP expression,revealing the crosstalk interaction between YAP/Notch regulatory signals.Based on the results of this model,it can be predicted that when there is crosstalk between YAP and Notch signals,YAP will induce Notch signal anomalies.Similarly,abnormal Notch signaling can also affect the YAP signaling pathway.In normal cellular processes,YAP and Notch dynamically synergistically regulate cell processes.This result can be used to predict the presence of lesions triggered by abnormal YAP and Notch signaling.The theoretical results obtained in this article are consistent with experimental observations,further revealing the physical mechanism of crosstalk between YAP and Notch signals,and providing valuable reference for further research on the crosstalk regulation of YAP and Notch signals in the future.
