摘要
以油胺为还原剂,十八烯和聚乙烯吡咯烷酮为分散剂,借助微流控技术合成了超小球形金纳米颗粒。通过紫外分光光度计(UV-Vis)、X-射线衍射(XRD)、能谱仪(EDX)、高分辨透射电子显微镜(HR-TEM)以及选区电子衍射(SAED)等分析,对超小金纳米颗粒的相结构、成分和形貌进行了全面表征。基于这些超小金纳米颗粒,通过工艺优化,成功制备出了急性心梗快速检测试剂盒,并对其性能进行考察。结果表明:合成的金纳米颗粒具有多晶结构,平均粒径仅4.57 nm;基于上述金纳米颗粒,获取的急性心梗快速检测试剂盒可对心梗进行快速检测,对心梗标志物(心肌肌球蛋白结合蛋白C)的检测限可低至0.1 ng/mL;同时,该试剂盒呈现出了高的特异性、重复性和稳定性,具有应用潜质。
Spherical Au nanoparticles(Au NPs)with ultrasmall size were controllably synthesized in oleylamine/octadecene/PVP system via microreation.The phase structure,composition and morphology of the Au NPs were comprehensively characterized by various analytical techniques,e.g.ultraviolet-visible spectroscopy(UV-Vis),X-ray diffraction(XRD),energy dispersive X-ray spectroscopy(EDX),high-resolution transmission electron microscopy(HR-TEM),and selected area electron diffraction(SAED).Based on these Au NPs,the acute myocardial infarction rapid test kit(AMIRTK)was successfully fabricated through process optimization and its performance was investigated in detail.It was found that as-synthesized Au NPs exhibited a polycrystalline structure and ultrasmall size(4.57±0.3 nm).Obtained AMIRTK with the above Au NPs could achieve the rapid detection and showed a very low limit of detection(0.1 ng/mL)for cardiac myosin binding protein-C(cMyBP-C)as myocardial marker.Meanwhile,the AMIRTK had high specificity,repeatability and stability,indicating a great potential in practice.
