摘要
“纸色谱法分离和鉴定氨基酸实验”作为分析化学实验的必修实验之一,具有操作简便,易于理解等优点,但也存在器材繁杂,实验装置利用率低等问题。本方案舍弃“纸色谱法分离和鉴定氨基酸”原实验中体积大、价格昂贵、重复使用率低的层析筒,选用较为简易、便宜易得的烧杯和保鲜膜作为层析装置,以及改进两种不同的展开模式供大家参考选择,并且将显色剂溶剂从有毒刺激性的正丁醇改为低毒的95%乙醇。改进的实验方案分离效果好可作为教学实验广泛开展,并且可以在课堂进行对比实验,提高学生的思考能力和探究能力。
“The Paper Chromatography Separation and Identification of Amino Acids Experiment”is one of the mandatory experiments in Analytical Chemistry,which has the advantages of simple operation and easy understanding.However,there are also problems such as complex equipment and low utilization of experimental equipment.The large volume,expensive and low reuse chromatography tube used in the Paper Chromatography Separation and Identification of Amino Acids experiment were abandoned,and a relatively simple,inexpensive and easily obtainable beaker and cling film was selected as the chromatography device,two different unfolding modes for everyone to refer to and choose from were improved,and the solvent in the chromogen was changed from toxic-stimulating n-butanol to low-toxic 95%ethanol.The good separation effect of the improved experimental scheme can be widely used as teaching experiments,and the comparative experiments can be carried out in class to improve students thinking ability and inquiry ability.
