摘要
采用细胞、透明质酸酶与人体临床实验,探讨积雪草提取物的舒缓与修护功效。细胞实验研究积雪草提取物对RAW264.7(小鼠巨噬细胞)分泌IL-6的影响,同时研究对HaCaT(人永生化表皮细胞)迁移的影响。通过抑制透明质酸酶活性评价积雪草提取物的舒缓效果。人体临床实验测试志愿者使用积雪草提取物精华液前后皮肤经皮水分流失TEWL值与皮肤含水量MMV值。积雪草提取物在0.003%、0.0015%时对IL-6浓度有明显抑制作用;体积浓度为0.2%、0.1%、0.05%的积雪草提取物能显著抑制透明质酸酶活性。100μg/mL的积雪草提取物对HaCaT细胞迁移有显著促进作用。人体临床实验表明精华液中添加0.1%的积雪草提取物与空白组相比,皮肤经皮失水TEWL值变化率为-37.86%(P<0.001),皮肤含水量MMV值变化率为65.09%(P<0.001)。
Cell experiment,hyaluronidase experiment and human clinical experiment were used to investigate the soothing and repairing effects of Centella asiatica extract.Cell experiments were conducted to study the effects of Centella asiatica extract on proliferation of RAW264.7 cells(mouse macrophages)and secretion of IL-6 in RAW cells,and the effects of Centella asiatica extract on migration of HaCaT cells(keratinocytes).Hyaluronidase assay was used to evaluate the soothing effect of Centella asiatica extract by inhibiting the activity of hyaluronidase.The human clinical experiment measured the skin water loss TEWL value and skin water content MMV value of volunteers before and after using Centella asiatica extract,and used VC20 image system to detect fine line improvement.The results showed that when the volume concentration of Centella asiatica extract was 0.003%and 0.0015%,the proliferation of RAW cells and IL-6 concentration were significantly inhibited.The activity of hyaluronidase was significantly inhibited by 0.2%,0.1%and 0.05%of the extracts.The extract of 100μg/mL could significantly promote the migration of HaCaT cells.Human clinical experiments showed that the change rate of skin transdermal water loss TEWL value was-37.86%(P<0.001),the change rate of skin water content MMV value was 65.09%(P<0.001).To sum up,Centella asiatica extract has certain soothing and repairing effects.
