摘要
目的基于一测多评法(QAMS)建立高效液相色谱法(HPLC)测定金菊五花茶颗粒中原儿茶酸、绿原酸、芦丁、山柰酚-3-O-芸香糖苷、射干苷、槲皮素、蒙花苷7种成分的含量。方法采用Thermo Hypersil GOLD a Q C_(18)(250 mm×4.6mm,5μm)色谱柱,以乙腈-0.1%磷酸溶液为流动相进行梯度洗脱,流速为1.0 ml/min,柱温为30℃,检测波长为264 nm,进样量为10μl。以芦丁为内参物,计算相对校正因子并进行耐用性考察,通过QAMS和外标法(ESM)计算7种成分的含量。结果对7种成分进行了方法学验证的考察,专属性、线性关系、精密度、重复性、稳定性均良好,验证结果符合要求。7种成分的平均加样回收率为96.86%~98.35%,相对标准偏差(RSD)为0.37%~1.40%。10批样品中均能检出7种成分,并且QAMS与ESM计算得到的含量的相对平均偏差(RAD)<2%。结论QAMS稳定可靠,可用于金菊五花茶颗粒中7种成分的含量测定。
Objective To establish a quantitative analysis of multi-components by single marker method(QAMS)for the simultaneous determination of Protocatechuic acid,Chlorogenic acid,Rutin,Kaempferol-3-O-rutoside,Tectoridin,Quercetin and Linarin in Jinju Wuhuacha Granule by High Performance Liquid Chromatography(HPLC).Methods The analysis was carried out on an analytical column Hypersil GoldAQ C_(18)(250 mm×4.6 mm,5μm)with gradient elution by acetonitrile and 0.1%phosphoric acid solution,flow speed was 1.0 ml/min,column temperature was 30℃,detection wavelength was 264 nm,and injection volume was 10μl.Using rutin as the internal reference,the relative correction factor was calculated and the durability was investigated,and the content of 7 components was calculated by QAMS and External Standard Method(ESM).Results The methodological verification of 7 components was investigated,and the specificity,linear relationship,precision,repeatability and stability were all good,and the verification results met the requirements.The average recoveries were 96.86%~98.35%and the relative standard deviations(RSD)were 0.37%~1.40%.Seven components were detected in all 10 batches of samples,and the relative mean deviation(RAD)between QAMS and ESM calculated concentrations was<2%.Conclusion QAMS is stable and reliable,and can be used for the determination of 7 components in Jinju Wuhua tea granules.
