摘要
目的:本研究旨在探究沉默HIF-1α对喉癌中SLC7A11的具体作用,为临床诊断喉癌寻找新的肿瘤标志物和寻找潜在治疗的靶点。方法:培养UMSCC5喉癌细胞,转染HIF-1α-siRNA,Western blot筛选最佳转染效率,针对转染后的细胞CCK-8检测增殖活性,流式细胞术检测各组细胞凋亡情况,Transwell法检测细胞侵袭和迁移,Western blot检测HIF-1α和SLC7A11表达。结果:成功沉默HIF-1α的表达,其中HIF-1α-siRNA1组的转染效率最高、最稳定(P<0.05)。沉默HIF-1α可以显著抑制人喉癌UMSCC5细胞的增殖(P<0.05)、促进细胞凋亡(P<0.05)以及抑制喉癌细胞侵袭、迁移(P<0.05)。沉默HIF-1α可以明显下调SLC7A11蛋白表达量(P<0.05)。结论:沉默HIF-1α可抑制人喉癌UMSCC5细胞增殖、侵袭、迁移行为以及凋亡,同时抑制SLC7A11的表达。为将来基于HIF-1α进行喉癌临床分子靶向治疗提供新的思路和理论依据。
Objective:To explore the specific effect of silencing HIF-1αon SLC7A11 in laryngeal cancer,and to find new tumor markers and potential therapeutic targets for clinical diagnosis of laryngeal cancer.Methods:UMSCC5 laryngeal cancer cells were cultured and transfected with HIF-1α-siRNA.Western blot was used to screen the best transfection efficiency.CCK-8 was used to detect the proliferation activity of the transfected cells.Flow cytometry was used to detect the apoptosis of each group.Transwell method was used to detect cell invasion and migration.Western blot was used to detect the expression of HIF-1αand SLC7A11.Results:The expression of HIF-1αwas successfully silenced,and the transfection efficiency of HIF-1α-siRNA1 group was the highest and the most stable(P<0.05).Silencing HIF-1αcan significantly inhibit the proliferation of human laryngeal carcinoma UMSCC5 cells(P<0.05),promote apoptosis(P<0.05),and inhibit the invasion and migration of laryngeal carcinoma cells(P<0.05).Silencing HIF-1αcould significantly down-regulate the expression of SLC7A11 protein(P<0.05).Conclusion:Silencing HIF-1αcan inhibit the proliferation,invasion,migration and apoptosis of human laryngeal carcinoma UMSCC5 cells,and inhibit the expression of SLC7A11.It provides new ideas and theoretical basis for clinical molecular targeted therapy of laryngeal cancer based on HIF-1αin the future.
