摘要
目的探究网状体家族基因1(Reticulon family gene1,RTN1)在结直肠癌(Colorectal cancer,CRC)组织中的表达及意义。方法通过利用TIMER、GEO和UALCAN数据库,对RTN1在结直肠癌及周围正常组织中的表达差异进行分析;应用GEPIA2在线分析工具分析RTN1在CRC中的表达模式和相关基因;借助STRING数据库挖掘与RTN1相互关联的蛋白;通过蛋白免疫印迹试验和免疫组化试验对以上生物信息学分析结果进行验证,以深入探索RTN1在CRC组织及周围正常组织中的差异表达。结果根据TIMER数据库的分析,RTN1在结直肠癌组织中的表达水平明显低于正常组织(均P<0.001)。GEO数据库中的四个数据集也表明,RTN1在CRC中的表达水平显著低于正常组织(均P<0.01)。UALCAN数据库进一步证实了这一结果,无论在哪种临床属性下,RTN1在CRC中的表达都低于正常组织。GEPIA2的分析结果表明,无论是结肠癌还是直肠癌,RTN1的表达均显著低于相邻的正常组织(均P<0.05),并且与RTN1表达呈正相关的四个基因(CSF1R、CD4、SLC9A9和ARHGEF6)在结直肠癌中也表现出类似的表达模式。STRING数据库的挖掘显示,RTN1可能与SPAST、REEP5、ATL3、ATL2、RTN4、ATL1、RTN2、TMEM33、BACE1和MANF等蛋白有交互作用。蛋白免疫印迹和免疫组化试验均证实,RTN1在CRC组织中的表达低于邻近正常组织。结论RTN1在结直肠癌组织中的表达低于癌旁正常组织,这一发现可能对CRC的诊断及治疗具有重要意义。
Objective To investigate the expression and significance of Reticulon family gene1(RTN1)in Colorectal cancer(CRC).Methods The expression difference of RTN1in CRC and its adjacent normal tissues was analyzed by TIMER,GEO and UALCAN databases.The expression pattern and related genes of RTN1in CRC tissues were studied by GEPIA2online analysis tool.The protein associated with RTN1was mined by STRING database.The above bioinformatic analysis results were verified by Western Blot test and Immunohistochemical test to further explore the differential expression of RTN1in CRC tissues and its adjacent normal tissues.Results Analysis from the TIMER database indicated that the expression level of RTN1was significantly reduced in CRC tissues compared to normal tissues(P<0.001).Analysis of four datasets from the GEO database also revealed that RTN1expression levels in colorectal cancer were dramatically diminished compared to normal tissues(P<0.01).This result was further confirmed by the UALCAN database,which showed that RTN1expression was lower in CRC than in normal tissue regardless of clinical attribute.Analysis using GEPIA2indicated that RTN1expression in both colon and rectal cancer was obviously decreased compared to the normal tissues(P<0.05),and the four genes positively correlated with RTN1expression(CSF1R,CD4,SLC9A9and ARHGEF6)also showed similar expression patterns in CRC.Mining the STRING database shows that RTN1 may interact with proteins such as SPAST,REEP5,ATL3,ATL2,RTN4,ATL1,RTN2,TMEM33,BACE1,and MANF.Both Western Blot and Immunohistochemical analyses verified that RTN1expression was lower in colorectal cancer tissues compared to normal tissues.Conclusion RTN1expression in colorectal cancer is found to be lower compared to that in the normal tissues,which may have important implications for the diagnosis and treatment of CRC.
