中药线香体外抑菌试验及血清代谢组学分析

摘要 目的:探究自制中药线香的体外抑菌效果、安全性及其对小鼠代谢的影响。方法:使用中药材料制作中药线香,采用牛津杯法比较自制中药线香与商品化线香燃烟溶液的体外抑菌效果,并通过气相色谱-质谱联用(GC-MS)检测中药线香燃烟中的抑菌成分。将24只雌性昆明小鼠随机分成空白对照组、中药线香熏蒸组和商品化线香熏蒸组,比较各组小鼠的肺脏和气管组织病变情况。收集血清样本,运用液相色谱-质谱联用(LCMS)非靶向代谢组学技术研究中药线香对小鼠血清代谢的影响。结果:体外抑菌结果显示,中药线香燃烟溶液对金黄色葡萄球菌和白色念珠菌的抑菌圈大于商品化线香燃烟溶液(P<0.05)。GC-MS检测结果显示,中药线香燃烟中含有具有广谱抑菌作用的活性成分,如α-松油烯(α-Terpinene)、薄荷醇(menthol)、β-石竹烯(β-Caryophyllene)和甲基庚烯酮(methylheptenone)。小鼠组织切片结果表明中药线香的安全性优于商品化线香。LC-MS结果显示,中药线香熏蒸组小鼠血清中的野芝麻新苷(Lamiide)、二氢硫辛酸(Dihydrolipoate)、对苯二酚(Hydroquinone)、L-谷氨酸(L-Glutamic acid)水平上调(P<0.05)。结论:自制中药线香具有良好的抑菌、抗炎、抗氧化的功效,且安全性较好。 Objective:To investigate the in vitro antibacterial efficacy,safety,and metabolic effects of homemade Chinese herbal incense on mice.Methods:Traditional Chinese medicinal materials were used to create the Chinese herbal incense.The in vitro antibacterial effects of the homemade Chinese herbal incense and commercial incense smoke solutions were compared using the Oxford cup method.Additionally,GC-MS analysis was employed to detect antibacterial components in the smoke of the Chinese herbal incense.Twenty-four female Kunming mice were randomly divided into three groups:a blank control group,a group exposed to Chinese herbal incense fumigation,and a group exposed to commercial incense fumigation.Lung and tracheal tissue lesions were assessed in each group of mice.The LC-MS untargeted metabolomicsis technology was used to study how the Chinese herbal incense affects the mice serum metabolism.Results:The in vitro antibacterial results demonstrated that the bacteriostatic zone diameter of the homemade Chinese herbal incense smoke solution against Staphylococcus aureus and Candida albicans was significantly larger than that of the commercial incense smoke solution(P<0.05).GC-MS analysis revealed that the smoke of the Chinese herbal incense contained active ingredients with a broad spectrum of antibacterial activity,such asα-Terpinene,menthol,β-Caryophyllene and methylheptenone.Tissue section analysis of mice showed that the safety of homemade herbal incense was superior to commercially available incense.LC-MS results indicated significantly upregulated serum levels of Lamiide,Dihydrolipoate,Hydroquinone,and L-Glutamic acid in the group exposed to Chinese herbal incense fumigation(P<0.05).Conclusion:The homemade herbal incense has excellent antibacterial,anti-inflammatory,and antioxidant effects,with good safty.
