盐驯化青花菜种子提高幼苗耐盐性的研究

摘要 通过研究盐驯化青花菜种子对提高其幼苗耐盐性的影响,为青花菜耐盐栽培提供理论支撑。本研究采用0、10、25、50、75、100 mmol/L NaCl对青花菜种子进行盐驯化7 d,以发芽指标为标准筛选出75、100 mmol/L为盐驯化浓度,测定其幼苗在高盐处理(225 mmol/L NaCl)下的光合色素含量、抗氧化酶活性及渗透调节物质含量。结果表明:盐驯化青花菜种子使其幼苗在高盐胁迫下的SOD、CAT活性及光合色素、脯氨酸和可溶性蛋白含量较CK2(盐对照)显著增加,可溶性糖和MDA含量显著下降。其中75 mmol/L驯化组与CK2相比,总叶绿素、叶绿素a、叶绿素b、类胡萝卜素显著提高283.95%、289.22%、277.13%、276.64%,SOD活性显著增加16.64%,脯氨酸含量显著增加120.76%,POD活性显著增加414.75%。综合分析表明,盐驯化青花菜种子可以提高其幼苗的耐盐性,隶属函数分析结果得出,75 mmol/L NaCl驯化青花菜种子的驯化效果最佳。 By studying the effect of salt domestication of broccoli seeds on improving their salt tolerance,theoretical support is provided for salt tolerant cultivation of broccoli.Broccoli seeds were acclimated to salt for 7 days with 0,10,25,50,75,and 100 mmol/L NaCl.Using germination indicators as criteria,75 and 100 mmol/L were selected as salt acclimation concentrations.Photosynthetic pigments,antioxidant enzyme activity,and osmoregulation substance content of their seedlings were measured under high salt treatment(225 mmol/L NaCl).The results showed that salt acclimation of broccoli seeds significantly increased SOD activity,CAT activity,the content of photosynthetic pigments,proline,and soluble protein and significantly decreased the content of soluble sugar and MDA under high salt stress compared with CK2(salt control).Compared with CK2,the 75 mmol/L domestication group showed a significant increase of 283.95%,289.22%,277.13%,and 276.64%in total chlorophyll,chlorophyll a,chlorophyll b,and carotenoids,with a significant increase of 16.64%in SOD activity,120.76%in proline content,and 414.75%in POD activity.Comprehensive analysis shows that salt domestication of broccoli seeds can improve their salt tolerance,and membership function analysis results show that 75 mmol/L NaCl domestication of broccoli seeds has the best domestication effect.
