铁皮石斛的HPLC指纹图谱

摘要 目的研究并建立不同产地铁皮石斛药材的指纹图谱。方法以流动相乙腈-0.4%(φ)磷酸溶液进行梯度洗脱,选用PRODIGY(4)C18(4.6 mm×250 mm,5μm)色谱柱,检测波长210 nm,流速1.0 mL/min,柱温30℃。以柚皮素峰为参照峰,在同一色谱条件下测定11批不同产地铁皮石斛药材,采用“中药色谱指纹图谱相似度评价系统(2012版)”对11批铁皮石斛进行相似度分析。结果建立了不同产地铁皮石斛药材的指纹图谱,并确认了10个共有特征峰,确认了新西兰牡荆苷2(1号峰)、夏佛塔苷(2号峰)、槲皮素(6号峰)、柚皮素(7号峰)4种成分;相似度评价结果显示,8批样品的相似度均大于0.900,其余3批相似度在0.782~0.877之间。结论本方法操作性强、稳定可靠、重复性好,符合指纹图谱分析的要求,为完善铁皮石斛药材的质量控制标准奠定了基础。 Objective To study and establish the fingerprints of Dendrobium officinale from different origins.Methods HPLC was conducted on Agilent PRODIGY(4)C18(4.6 mm×250 mm,5μm).Cetonitrile-0.4%phosphoric acid aqueous solution was employed as mobile phase with gradient elution,the detection wavelength was set at 210 nm,the flow rate was 1.0 min/mL,and the column temperature was maintained at 30℃.With naringenin as reference substance,the fingerprints of 11 batches of Dendrobium officinale were analyzed under the same chromatographic conditions.The Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia Media(version 2012)was used to analyze the similarity of 11 batches of Dendrobium officinale.Results The fingerprints of Dendrobium officinale from different origins were established,10 common characteristic peaks were confirmed,and 4 components were identified,namely viceninII(peak 1),schafloside(peak 2),quercetin(peak 6)and naringenin(peak 7).The similarity results showed that the 8 batches of Dendrobium officinale was more than 0.900,and the remaining medicinal materials were similar within the range from 0.782 to 0.877.Conclusion The method has high operability,strong reliability and good reproducibility,which conforms the requirements of fingerprint analysis,and lays a research foundation for improving the quality control standards of Dendrobium officinale.
