摘要
旨在探究冷冻稀释液中添加不同浓度的瘦素(LEP)对豫西黑猪精子冷冻保存效果的影响。选用12头豫西黑公猪精液,在冷冻稀释液中添加浓度分别为0、2.5、5、7.5、10 ng/mL的LEP,检测解冻后豫西黑猪精子的运动参数、功能参数、受孕率等指标,筛选出最佳LEP添加浓度;用最佳浓度组和对照组精液分别对发情母猪人工授精,观察受孕效果。结果显示:冷冻稀释液中添加5 ng/mL LEP组与其他组在精子的活力、曲线速度(VCL)、直线速度(VSL)、平均路径速度(VAP)、线性度(LIN)方面差异显著(P<0.05);在线性指数(STR)方面,5、7.5、10 ng/mL LEP组之间差异不显著(P>0.05),但均与对照组差异显著(P<0.05);在振动指数(WOB)方面,5 ng/mL LEP组与其他各组差异显著(P<0.05);在DNA完整率、质膜完整率、线粒体膜电位和顶体完整率方面,5 ng/mL LEP组与其他各组差异显著(P<0.05);新鲜精液组与5 ng/mL LEP组受孕率结果相近(P>0.05)。由此可见,在冷冻稀释液中添加LEP能有效提高豫西黑猪精子的冷冻效果,可能是由于LEP减轻了精子在快速冷冻和解冻过程中带来的氧化应激损伤,提高了精子冷冻效果和受孕率。
This experiment was conducted to investigate the effects of different concentrations of leptin(LEP)added to frozen liquid on the cryopreservation of sperm from Yuxi black swine.The semen of 12 Yuxi black boars was added with the concentrations of 0,2.5,5,7.5 and 10 ng/mL LEP in the frozen dilution.Then,the motility parameters,functional parameters and conception rate of the sperm were detected after thawing,and the optimal concentration of LEP was selected.The semen of the optimum concentration group and the control group were used for artificial insemination of estrus sows,and the fertilization effects were observed.The results showed as follows:There were significant differences in sperm motility,VCL,VSL,VAP and LIN between the 5 ng/mL LEP group and the other groups(P<0.05).In terms of STR,there was no significant difference among the 5,7.5 and 10 ng/mL LEP groups(P>0.05),but there was significantly difference from the control groups.In terms of WOB,the 5 ng/mL LEP group was significantly different from the other groups(P<0.05).In terms of DNA integrity rate,plasma membrane integrity rate,mitochondrial membrane potential and acrosome integrity rate,there were significant differences between the 5 ng/mL LEP group and the other groups(P<0.05).The results of pregnancy rate were similar between the fresh semen group and the 5 ng/mL LEP group(P>0.05).In conclusion,adding LEP to the freezing base fluid effectively improved the freezing effect of Yuxi black swine sperm,which might be the reason why LEP reduces the oxidative stress damage caused by sperm in the process of rapid freezing and thawing;and the addition improved the freezing effect of sperm and raised the pregnancy rate in the black swine.
