摘要
针对多粘类芽孢杆菌发酵工艺进行系统优化,主要从培养基成分(碳源、氮源、碳酸钙)方面入手,通过三角瓶试验及100 L发酵罐放大试验找出最优发酵培养基,提高多粘类芽孢杆菌的菌量和芽孢率。结果表明,最佳发酵培养基为2%豆粕粉+2%蔗糖+1%玉米淀粉+0.6%硫酸铵,最优碳酸钙添加量为0.5%,在37℃、转速200 r/min、通气60 L/min、初始pH值7.2的条件下,100 L发酵罐发酵32 h,菌量和芽孢率达到最佳,验证了小试条件的可行性和稳定性。
The study systematically optimizes the fermentation process of Bacillus Polymyxoides from the main aspects of medium composition(carbon source,nitrogen source,calcium carbonate)and fermentation conditions(pH value,inoculation amount,liquid loading amount,rotational speed and temperature),etc.;and finds out the optimal fermentation conditions through triangle bottle test and 100 L fermenter amplification test,so as to improve the bacteria quantity and spore rate of Bacillus Polymyxoides.The results show that the optimal fermentation medium is 2%soybean meal meal+2%sucrose+1%corn starch+0.6%ammonium sulfate,and the optimal calcium carbonate addition is 0.5%.Under the conditions of 37℃,200 r/min,60 L/min ventilation and initial pH value of 7.2,the fermentation in 100 L fermenter for 32 h achieves the best bacterial content and spore rate.This verifies the feasibility and stability of the experimental conditions.
