摘要
目的建立全自动碘分析仪检测尿碘的方法,与砷铈催化分光光度法(下称手工法)进行比较,分析该方法在0~600μg/L范围内检测尿碘结果的可靠性、稳定性以及替代手工法的可行性。方法采用全自动碘分析仪检测0~600μg/L范围内尿碘,验证其检出限、相关系数、精密度、准确度、加标回收率等方法学指标,并与手工法同时检测尿液标本中尿碘含量,对测定结果进行比对。结果在标准系列0~600μg/L浓度范围内,相关系数r=0.9998,方法检出限为0.87μg/L,低、中、高三个浓度的尿碘有证标准物质检测结果均在证书规定范围内,相对标准偏差分别为2.29%、0.95%、0.60%,平均加标回收率为99.00%、100.93%、99.76%。与手工法同时测定30份尿液标本,二者结果比较差异无统计学意义(t=-0.618,P=0.542)。结论全自动碘分析仪法精密度和准确度高,线性范围宽,检测效率高,在0~600μg/L范围内可以替代手工法,适合推广应用。
Objective To establish a method for detecting urinary iodine by automatic iodine analyzer,to compare it with arsenic cerium catalytic spectrophotometry(manual method),and to analyze the reliability and stability of the method in the range of 0-600μg/L,and the feasibility of replacing the manual method.Methods Automatic iodine analyzer was used to detect urinary iodine in the range of 0-600μg/L,and the detection limit,correlation coefficient,precision,accuracy,standard addition recovery rate and other methodological indicators were verified.The urinary iodine content in urine samples was detected at the same time with manual method,and the results were compared.Results In the standard series of 0-600μg/L concentration range,the correlation coefficient r=0.9998,the detection limit of the method was 0.87μg/L,the low,medium and high concentrations of urinary iodine certified reference materials detection results were within the range of the certificate,the relative standard deviation were 2.29%、0.95%、0.60%,respectively.The average recoveries were 99.00%,100.93%and 99.76%.There was no significant difference in the results of 30 urine samples measured simultaneously with the manual method(t=-0.618,P=0.542).Conclusion The automatic iodine analyzer method has high precision and accuracy,wide linear range,high detection efficiency,and can replace the manual method in the range of 0-600μg/L,which is suitable for popularization and application.
