摘要
目的探讨IL18RAP基因与肝癌患者预后和肿瘤微环境中CD8^(+)T细胞浸润的相关性以及作为肿瘤标志物的可能性。方法癌症基因组图谱(The Cancer Genome Atlas,TCGA)用于评估IL18RAP基因在肝癌中的表达。单因素Cox分析、多因素COX分析和生存分析揭示IL18RAP基因的预后价值。KEGG、GO和Hallmark富集分析寻找与IL18RAP基因相关的功能通路。免疫浸润分析探究IL18RAP基因与22种免疫细胞浸润的关系。通过单细胞测序数据库与免疫组化验证IL18RAP基因与CD8^(+)T细胞浸润的相关性。结果IL18RAP在肝癌组织中表达下调,其低表达与肝癌患者的不良预后相关。功能富集分析显示IL18RAP的表达与免疫功能通路相关。免疫浸润、单细胞测序和免疫组化表明IL18RAP高表达于CD8^(+)T细胞。结论IL18RAP低表达与肝癌患者的不良预后相关,可能影响了抗肿瘤免疫的CD8^(+)T细胞。
Objective To investigate the correlation between the IL18RAP gene and the prognosis of liver cancer patients and CD8^(+)T cell infiltration in tumor microenvironment,and the possibility of IL18RAP as a tumor biomarker.Methods The Cancer Genome Atlas(TCGA)was used to evaluate the expression of IL18RAP in liver cancer.Univariate Cox analysis,multivariate COX analysis,and survival analysis revealed the prognostic value of the IL18RAP.KEGG,GO,and Hallmark enrichment analyses were conducted to identify functional pathways associated with the IL18RAP.Immune infiltration analysis was conducted to explore the relationship between the IL18RAP gene and 22 types of immune cell infiltration.The correlation between IL18RAP and CD8^(+)T cell infiltration was verified by single-cell sequencing database and immunohistochemistry.Results IL18RAP is downregulated in liver cancer tissues and its low expression is associated with poor prognosis in patients with liver cancer.Functional enrichment analysis showed that the expression of IL18RAP is related to immune function pathways.Immune infiltration,single-cell sequencing,and immunohistochemistry showed that IL18RAP was highly expressed in CD8^(+)T cells.Conclusion Low expression of IL18RAP is associated with poor prognosis in liver cancer patients,which may affect the anti-tumor immune of CD8^(+)T cells.
