Prognostic value of SNHG12 in bladder urothelial carcinoma

目的lncRNA SNHG12分子在各种恶性肿瘤的增殖和侵袭过程中起着至关重要的作用,通过收集数据进行分析,评估SNHG12在膀胱尿路上皮癌(BUC)预后评估的临床意义。方法采用qRT-PCR技术评估40例配对BUC组织和邻近非癌组织(ANCTs)中SNHG12的表达。并与患者临床特征进行单因素以及多因素分析,采用SNHG12表达量高于50%作为高表达组,低于50%作为低表达组,构建Kaplan-Meier生存曲线研究高表达组和低表达组无复发生存率(recurrence free survival,RFS)的差异,采用受试者工作特征(ROC)曲线分析SNHG12在BUC中的预后评估的临床价值。通过qPCR检测SNHG12在膀胱尿路上皮癌细胞5637和正常膀胱黏膜细胞SV-HUC-1的表达差异,转染siRNA和(或)质粒的5637细胞检测膀胱尿路上皮癌细胞增殖、迁移和侵袭能力。结果与ANCTs相比,在BUC标本中观察到SNHG12表达水平明显升高(P<0.001)。且SNHG12的高表达与肿瘤大小(P=0.01)和复发(P=0.003)有关。多变量分析表明,SNHG12是BUC的独立预后因素。SNHG12表达量高的患者的平均RFS月数短于SNHG12表达量低的患者(P=0.0167),ROC曲线分析结果显示,SNHG12的ROC曲线下面积(AUC)值为0.8200(95%CI:0.7182~0.9218,P<0.001)。经过细胞qPCR结果显示,SNHG12在5637细胞和SV-HUC-1中表达差异有统计学意义(P<0.05),CCK-8、Transwell和Matrigel试验表明SNHG12差异表达影响肿瘤细胞的增殖、迁移和侵袭能力。结论SNHG12有可能是一种独特的、潜在的BUC预后评估的生物标志物。

Objective Long non-coding RNA(lncRNA)SNHG12 plays a crucial role in the proliferation and invasion of various malignancies.This study aims to assess the clinical significance of SNHG12 in the prognostic evaluation of bladder urothelial carcinoma(BUC).Methods The authors evaluated SNHG12 expression in 40 paired BUC tissues and adjacent non-cancerous tissues(ANCTs)using qRT-PCR.Univariate and multivariate analyses were performed in relation to the patients′clinical characteristics.Kaplan-Meier survival curves were constructed to study the difference in recurrence-free survival(RFS)between high and low expression groups,and ROC curves were used to analyze the prognostic value of SNHG12 in BUC.The differential expression of SNHG12 in BUC cells(5637)and normal bladder mucosal cells(SV-HUC-1)was also assessed by qPCR,and the effect of SNHG12 on cell proliferation,migration,and invasion was tested using siRNA transfection and plasmid overexpression in 5637 cells.Results SNHG12 expression was significantly elevated in BUC samples compared to ANCTs(P<0.001).High SNHG12 expression was correlated with tumor size(P=0.01)and recurrence(P=0.003).Multivariate analysis indicated that SNHG12 is an independent prognostic factor for BUC.Patients with high SNHG12 expression had a shorter mean RFS compared to those with low SNHG12 expression(P=0.0167).The ROC analysis revealed that the AUC value of SNHG12 was 0.8200(95%CI:0.7182 to 0.9218;P<0.001).qPCR results showed a statistically significant difference in SNHG12 expression between 5637 and SV-HUC-1 cells,and CCK-8,Transwell,and Matrigel assays demonstrated that differential expression of SNHG12 affected the proliferation,migration,and invasion abilities of tumor cells.Conclusion These findings suggest that SNHG12 may be a unique and potential biomarker for the prognostic evaluation of BUC.
