摘要
目的:探究紫草素联合顺铂对人非小细胞肺癌细胞凋亡的作用研究,为临床合理用药提供科学依据。方法:将NCI-H2347细胞设空白对照组、紫草素(1μg/mL)组、顺铂组(50μg/mL)和联合实验组(紫草素1μg/mL+顺铂5μg/mL),MTT筛选出紫草素的使用浓度及联合用药后对NCI-H2347细胞的抑制作用;Western blot检测特异性蛋白TTF-1(甲状腺转录因子-1)及Bax与Bcl-2;凋亡试剂盒进一步检测Bax与Bcl-2。结果:MTT结果显示紫草素的有效浓度为1μg/mL;Western blot法检测NCL-H2347细胞中紫草素联合顺铂组TTF-1较单独使用紫草素或顺铂组明显降低;紫草素联合顺铂能明显增加Bax与Bcl-2的比值。结论:紫草素联合顺铂后对TTF-1的抑制可能与凋亡途径有关。
Objective:Exploring the effect of purpurin combined with Cisplatin on apoptosis of human non-small cell lung cancer cells,providing scientific basis for rational clinical medication.Method:NCI H2347 cells were divided into a blank control group,a Shikonin group(1μg/mL),a Cisplatin group(50μg/mL),and a combination(Shikonin 1μg/mL+Cisplatin 5μg/mL)experimental group.MTT was used to screen the concentration of Shikonin and the inhibitory rate of the combination on NCI H2347 cells;Western blot detection of specific protein TTF 1(thyroid transcription factor 1)and Bax and Bcl 2;Further detection of Bax and Bcl 2 using apoptosis assay kit.Result:The MTT results showed that the effective concentration of shikonin was 1μg/ml;Western blot analysis showed that the TTF 1 levels in NCL H2347 cells were significantly reduced in the combination of shikonin and cisplatin compared to those treated with shikonin or cisplatin alone;The combination of shikonin and cisplatin can significantly increase the ratio of Bax to Bcl 2.Conclusion:The inhibition of TTF 1 by the combination of shikonin and cisplatin may be related to the apoptotic pathway.
