摘要
目的:通过构建单纯疱疹病毒性角膜炎(HSK)小鼠模型,观察银翘散对HSK小鼠TNF-α和IFN-γ表达的影响。方法:将120只小鼠随机平均分为A组、B组、C组,A组为正常对照组,B组为模型组,C组为药物组,B组、C组均进行HSK小鼠建模,C组使用0.1mL/10g体重的银翘散进行灌胃,B组小鼠使用0.1mL/10g体重的生理盐水进行灌胃,干预时间共为14天,A组小鼠不作干预。观察银翘散对HSK小鼠TNF-α和IFN-γ表达的影响。结果:造模第1天,B组和C组的TNF-α水平显著高于A组,A组、B组和C组的IFN-γ水平无统计学差异(P>0.05);造模第3天与造模第5天,B组和C组的TNF-α水平、IFN-γ水平显著高于A组(P<0.05),C组的TNF-α水平、IFN-γ水平显著高于B组(P<0.05);造模第7天,B组的TNF-α水平显著高于A组(P<0.05),C组的TNF-α水平显著低于A组(P<0.05),B组和C组的IFN-γ水平显著高于A组(P<0.05),C组的IFN-γ水平显著高于B组(P<0.05),C组的TNF-α水平显著低于B组(P<0.05);造模第14天,B组和C组的TNF-α水平显著低于A组(P<0.05),IFN-γ水平显著高于A组(P<0.05),C组的TNF-α水平显著低于B组(P<0.05),IFN-γ水平显著高于B组(P<0.05)。结论:银翘散可降低小鼠血清TNF-α水平,升高血清IFN-γ水平,调节细胞因子的平衡,但疗程较长,需坚持使用。
Objective:To observe the effect of Yinqiao Powder on the expression of TNF-α and IFN-γ in mice with herpes simplex keratitis(HSK)by constructing a mouse model.Methods:120 mice were randomly divided into group A,group B and group C.Group A was the normal control group,group B was the model group,group C was the drug group,and group B and group C all established HSK mice models.Group C was given Yinqiao Powder with 0.1mL/10g body weight,and group B mice were given normal saline with 0.1mL/10g body weight.The intervention time was 14 days.Group A was not intervened.The effect of Yinqiao powder on the expression of TNF-α and IFN-γ in HSK mice was observed.Results:On the first day of modeling,the levels of TNF-α in group B and group C were significantly higher than those in group A,while there was no statistical difference in IFN-γ levels among group A,group B and group C(P>0.05).On the 3rd and 5th day of modeling,the levels of TNFα and IFN-γ in groups B and C were significantly higher than those in group A(P<0.05),and the levels of TNF-α and IFN-γ in group C were significantly higher than those in group B(P<0.05).On the 7th day of modeling,the level of TNF-α in group B was significantly higher than that in group A(P<0.05),the level of TNF-α in group C was significantly lower than that in group A(P<0.05),and the levels of IFNγin groups B and C were significantly higher than that in group A(P<0.05).The level of IFN-γ in group C was significantly higher than that in group B(P<0.05),while the level of TNFαin group C was significantly lower than that in group B(P<0.05).On the 14th day of modeling,the levels of TNF-α in group B and C were significantly lower than those in group A(P<0.05),while the levels of IFN-γ in group B and C were significantly higher than those in group A(P<0.05).TNF-α in group C were significantly lower than those in group B(P<0.05)and IFN-γ higher than those in group B(P<0.05).Conclusion:Yinqiao Powder can decrease the serum TNF-α level,increase the serum IFN-γ level and adjust the balance of cytokines in mice,but the course of treatment is long,so it needs to be used persistently.
