摘要
肿瘤相关巨噬细胞(tumour-associated macrophages,TAMs)是肿瘤中常见的基质细胞类型之一,其促肿瘤M2亚型与肿瘤微环境的免疫抑制状态密切相关,并促进肿瘤的恶性进展。脂质代谢重编程是癌症进展的新特征,肿瘤细胞为了维持自身生存,会进行代谢转化以利用各种途径获得脂质和增加脂质氧化。相对的,TAMs的脂代谢也被重新编程,并参与调节其自身的极化和相应的功能与表型。研究表明,在肿瘤微环境中肿瘤进展过程促使TAMs中的脂质积累,导致TAMs极化为M2表型。全文阐明脂质代谢重编程下的TAMs的分化及其在支持癌症进展方面起到的重要作用,靶向干扰TAMs脂质代谢重编程或许可以为胃肠道肿瘤治疗提供新的治疗策略。
Tumor associated macrophage(TAMs)is the the most common stromal cell type in malignant tumors,and its M2 subtype is closely related to the immunosuppression of the tumor microenvironment to promote tumor progression.Tumor cells undergo metabolic transformation to obtain lipids and increase lipid oxidation through various pathways in order to maintain the survival.On the other hand,the lipid metabolism of TAMs is also reprogrammed and involved in regulating their polarization,and corresponding functions and phenotyping.Research has shown that in the tumor microenvironment the progression of tumor promotes lipid accumulation in TAMs,leading to polarization into M2 phenotype.This paper elucidates the differentiation of TAMs under lipid metabolism reprogramming and their role in supporting cancer progression.Gastrointestinal cancers are most common malignant tumors with high incidence and mortality,targeted interference with lipid metabolism reprogramming of TAMs may provide new therapeutic strategies for gastrointestinal tumors.
