高效液相色谱法同时测定护肝宁片中虎杖苷和白藜芦醇含量研究

Simultaneous Determination of the Content of Polydatin and Resveratrol in HuGanNing Tablets by HPLC

目的建立高效液相色谱法测定护肝宁片中白藜芦醇和虎杖苷含量的方法。方法色谱条件:色谱柱为安捷伦XDB-C_(18)(4.6×250 mm,5μm),流动相:乙睛∶水(A∶B)梯度洗脱(0~30 min:A 22%~40%,B 88%~60%;30~40 min:A 40%~22%,B 60%~88%);检测波长:306 nm;流速:1.0 mL·min^(-1);进样量:10μL。结果虎杖苷在2.2298~22.2977μg·mL^(-1)的范围内呈良好的线性关系(r=0.99999),平均回收率=103.1%,RSD=1.7%;白藜芦醇在1.0008~10.008μg·mL^(-1)的范围内呈良好的线性关系(r=0.9999),平均回收率=99.7%,RSD=1.2%。结论本测定方法简便可行,重复性好,可用于护肝宁片的质量控制。

OBJECTIVE To establish a HPLC method for the determination of resveratrol and polydatin in Huganning tablets.METHODS Chromatographic conditions:chromatographic column was Agilent XDB-C _(18)(4.6×250 mm,5μm),mobile phase∶acetonitrile∶water(A∶B)gradient elution(0-30 min:A 22%-40%,B 88%-60%;30-40 min:A 40%-22%,B 60%-88%);detection wavelength:306 nm;flow rate:1.0 mL·min^(-1);injection volume:10μL.RESULTS Polydatin in the range of 2.2298-22.2977μg·mL^(-1) showed a good linear relationship(r=0.99999),the average recovery was 103.4%,RSD=1.5%;Resveratrol showed a good linear relationship in the range of 1.0008-10.008μg·mL^(-1)(r=0.9999),the average recovery was 99.7%,RSD=1.8%.CONCLUSION The method is simple,feasible and reproducible,and can be used for the quality control of HuGanNing tablets.
