摘要
目的研究中药单体红景天苷对甲状腺功能减退大鼠的抗抑郁作用机制。方法将大鼠随机分为正常组、模型组、低剂量组、高剂量组和甲状腺素组,每组5只,除正常组外,其余各组大鼠采用丙基硫氧嘧啶(PTU)法构建甲状腺功能减退模型。造模成功后,甲状腺素组予优甲乐4.0μg/(kg·d),低、高剂量组分别予红景天苷15.0,30.0mg/(kg·d),灌胃28d。分别于给药前后对大鼠进行行为学评估(强迫游泳实验、糖水偏好度实验)和甲状腺激素(T_(3)、T_(4)、TSH)检测,并于给药后检测大鼠血清5-HT及脑组织炎症因子(IL-6、IL-1β、TNF-α)含量。结果与模型组(阴性对照)比较,红景天苷低、高剂量组大鼠抑郁行为显著改善,即强迫游泳不动时间均显著缩短(P<0.05或P<0.01),糖水偏好度均显著增多(P<0.01);甲状腺激素水平显著改善,即血清T_(3)、T_(4)浓度显著升高(P<0.05或P<0.01),血清TSH浓度显著下降(P<0.01);血清5-HT浓度显著升高(P<0.05或P<0.01);脑组织炎症损伤显著减轻,即脑组织Il-6、IL-1β及TNF-α含量均显著下降(P<0.01)。与甲状腺素组(阳性对照)比较,红景天苷高剂量组在改善甲状腺激素水平和减轻脑组织炎症损伤方面,差异无统计学意义(P>0.05),并能显著提升血清5-HT浓度(P<0.05)。结论红景天苷对甲状腺功能减退大鼠的抗抑郁作用是通过改善甲状腺激素水平,减轻脑组织炎症损伤,提高血清5-HT浓度而实现的。
Objective To study the mechanism of anti-depression effect of salidroside on hypothyroidism rats.Methods The rats were randomly divided into normal group,model group,low-dose group,high-dose group and thyroxine group,with 5 in each group.Except the normal group,the other groups were given propyl thiouracil(PTU)to construct the hypothyroidism model.Since the model was established,the rats in thyroid group were given Euthyrox of 4.0μg/(kg·d),while the low-dose and high-dose groups were given salidroside of 15.0 and 30.0mg/(kg·d)respectively,with intragastric administration for 28 days.The behavioral assessment,including forced swimming test and sucrose preference test and thyroid hormones,involving T3,T4 and TSH were measured before and after administration.The concentration of 5-HT in serum and inflammatory factors,concerning IL-6,IL-1βand TNF-αin brain tissue were detected after administration.Results Compared with the model group(negative control),the depression behavior of the rats in the low and high dose salidroside groups was significantly improved,that the forced swimming immobility time was significantly shortened(P<0.05 or P<0.01),while the sucrose preference was significantly increased(P<0.01),the thyroid hormone level was significantly improved,that the serum concentrations of T3 and T4 were significantly increased(P<0.05 or P<0.01),while that of TSH was significantly decreased(P<0.01),the serum concentrations of 5-HT was significantly increased(P<0.05 or P<0.01),and the inflammatory injury of brain tissue was significantly alleviated,that the contents of Il-6,IL-1βand TNF-αin brain tissue were significantly decreased(P<0.01).Compared with the thyroxine group(positive control),the high-dose salidroside group had no significant difference in improving thyroid hormone level and reducing inflammatory damage of brain tissue(P>0.05),and significantly increased serum 5-HT concentration(P<0.05).Conclusion The anti-depression effect of salidroside on hypothyroidism rats is achieved by improving thyroid hormone levels,reducing inflammatory damage of brain tissue,and increasing serum 5-HT concentration.
