孟德尔随机化分析蚕豆摄入量与帕金森病发病风险的因果关系

摘要 目的应用两样本孟德尔随机化方法(Mendelian randomization,MR)探究蚕豆摄入量与帕金森病发病风险之间的潜在因果关系。方法使用全基因组关联研究(genome-wide association study,GWAS)的汇总数据,选取与蚕豆摄入量显著相关的单核苷酸多态性(single nucleotide polymorphisms,SNPs)对蚕豆摄入量和帕金森病发病风险的因果效应进行孟德尔随机化分析,逆方差加权法(inverse-variance weighted,IVW)作为主要的分析方法,并采用多种方法进行敏感性分析来评估结果的稳定性和可靠性。结果逆方差加权法结果显示蚕豆摄入量与帕金森病(OR=0.966,95%CI:0.938~0.996,P=0.026)存在显著相关性。敏感性分析结果显示不存在基因的水平多效性及异质性(P>0.05)。结论两样本孟德尔随机化研究分析结果显示,蚕豆摄入量是帕金森病的保护因素,为帕金森病的治疗和预防提供了方向。 Objective To explore the causal relationship between broad bean intake and risk of Parkinson’s disease using a two-sample Mendelian randomization method.Methods Using pooled data from genome-wide association studies,single nucleotide polymorphisms significantly associated with broad bean intake were used as instrumental variables to evaluate the causal effect of broad bean intake with Parkinson’s disease,inverse-variance weighted was used as the main analysis method and the MR-Egger regression analysis and weighted median method were used for sensitivity analysis to evaluate the stability and reliability of the results.Results Inverse variance weighted method(OR=0.966,95%CI:0.938~0.996,P=0.026)showed causal relationship between broad bean intake and Parkinson.Sensitivity analysis showed no heterogeneity or horizontal pleiotropy of genes(P>0.05).Conclusion The results of the two-sample Mendelian randomization study showed a causal relationship between broad bean intake and Parkinson’s disease risk,providing a direction for the treatment and prevention of Parkinson’s disease.
