摘要
玉米是世界上最主要的农作物之一,深受农民的喜欢。玉米种子萌发质量关系着玉米的苗势、产量和品质,研究与玉米萌发相关的分子机制对于提高玉米种子的萌发质量有重要意义。本研究以玉米自交系537A为试验材料,采用miRNA测序技术,从全基因组角度鉴定与玉米萌发相关的miRNAs及靶基因,结果分别在2个比较组Y-53h vs Y-6h和Y-72h vs Y-53h共鉴定得到432和371个具有表达差异的miRNAs。对靶基因进行预测并分析表达量,结果发现靶基因表达量差异显著,参与了玉米种子的萌发过程。选取靶基因Zm00001d014991构建植物表达载体pBI121-Zm00001d014991转化拟南芥,发现拟南芥的萌发能力提高。本研究结果为深入研究玉米种子萌发的分子机制提供了参考。
Maize is one of the most important crops in the world and is very popular with farmers.The germination quality of maize seeds is related to the seedling potential,yield and quality of maize.It is of great significance to study the molecular mechanism related to maize germination to improve the germination quality of maize seeds.In this study,maize inbred line 537A was used as experimental material,and miRNA sequencing technology was used to identify miRNAs and target genes related to maize germination from the perspective of whole genome.A total of 432 and 371 differentially expressed miRNAs were identified in Y-53h vs Y-6h and Y-72h vs Y-53h,respectively.The expression levels of target genes were predicted and analyzed and the results showed that the expression levels of target genes were significantly different and involved in the germination process of maize seeds.The target gene Zm00001d014991 was selected and used to construct the plant expression vector pBI121-Zm00001d014991 to transform Arabidopsis thaliana,and the results showed that the germination ability of Arabidopsis thaliana was improved.The results provided a reference for further study of the molecular mechanism of maize seed germination.
