摘要
为深入探讨MATE(Multidrug and toxic compound extrusion)转运蛋白在百合花青素苷转运中的作用机制,本研究克隆了亚洲百合‘Curitiba’的DTX35基因,并和亚洲百合‘Tiny Padhye’的LhDTX35基因进行了比较分析。结果表明,两个亚洲百合品种的DTX35基因相似性很高,达到83.98%;都编码507个氨基酸,属于MATE转运蛋白;两个品种中DTX35基因编码的蛋白质的亲缘关系很近;大部分氨基酸具有疏水性;都具有12个跨膜α-螺旋结构;DTX35基因的表达概况和花瓣花青素苷含量呈正相关。研究结果表明两个亚洲百合‘Curitiba’和‘Tiny Padhye’的DTX35基因高度保守。两个品种中的DTX35基因功能相似,具备MATE转运蛋白特性。预示着亚洲百合‘Curitiba’的DTX35基因可能也参与相应花瓣的花青素苷的转运。本研究结果对深入解析百合花青素苷转运机制奠定理论基础,为百合花色分子育种提供基因资源。
To investigate the mechanism of the role of MATE(Multidrug and toxic compound extrusion)transporter in anthocyanin transportation in Lilium spp.,DTX35 gene was cloned in'Curitiba'of Asiatic hybrid lily.The LhDTX35 genes between the'Curitiba'and'Tiny Padhye'were compared.The results showed that DTX35 gene of'Curitiba'showed high similarity with that of'Tiny Padhye',reaching to 83.98%.Both encoded 507 amino acids,belonging to MATE transporter.The proteins encoded by DTX35 gene of two varieties are closely related phylogenetically;most amino acids of the proteins are hydrophobicity;they possessed 12 transmembraneα-helical structures;the expression profiles were positive related to content of anthocyanin of tepal.The results indicated that DTX35 genes were highly conservative.Functions of DTX35 gene were similar in two varieties,which possessed the characteristics of the MATE transport protein.It suggested that the DTX35 gene may be also involved in the transportation of anthocyanin of tepal in'Curitiba'of Asiatic hybrid lily.The work lay the theory basis on revealing the anthocyanin transport mechanism in Lilium and provided the new gene resources in molecular breeding of coloration in Lilium.
