摘要
为了筛选出茶树不同品种间分类所需要的相关DNA条形码引物。本研究用源于其他文献的matK、T-ITS、trnH-psbA等序列的通用引物及自行设计的rbcL引物,对茶树进行相关序列多样性分析。用试剂盒提取基因组总DNA,备选引物进行PCR扩增,琼脂糖凝胶电泳检测PCR扩增产物。结果显示,从matK、rcbL、T-ITS、trnH-psbA、rbcL这5对引物中,初步筛选出2对适合茶树DNA条形码的引物,分别是matK、rcbL,通过分析认为可以作为茶树DNA条形码的备选序列,为茶树的品种分类及种质资源研究提供一定的参考。
In order to screen out relevant DNA barcode primers needed for classification among different varieties of tea plants.In this study,the common primers of matK,T-ITS,trnH-psbA from other literatures and the self-designed rbcL primers were used to analyze the relevant sequence diversity of tea plants.The total DNA of the genome was extracted with the kit,the alternative primers were amplified by PCR,and the PCR amplification products were detected by agarose gel electrophoresis.The results showed that from the 5 pairs of primers matK,rcbL,T-ITS,trnH-psbA and rbcL,2 pairs of primers suitable for tea plant DNA barcode were preliminatively screened,namely mat K and rcbL,which could be considered as alternative sequences for tea plant DNA barcode through analysis.It provides some reference for the study of tea variety classification and germplasm resources.
