摘要
目的探究A型肉毒毒素(BONT/A)对坐骨神经损伤模型以及受损神经周围血管与雪旺细胞(SCs)的影响。方法将125只小鼠随机分为正常组(NC)、对照组(Control)以及BONT/A组,对照组以及BONT/A组(6U·kg^(-1))又各分为7d组和21d组,每组25只。对照组和BONT/A组通过挤压法建立坐骨神经损伤模型,正常组不做处理。观察小鼠整体状态并用坐骨神经功能指数(SFI)评估其运动功能。髓鞘甲苯胺蓝(TB)染色观测坐骨神经再生修复情况。Western blot检测SCs标志物神经生长因子受体p75(p75NTR)、髓磷脂蛋白零(MPZ)以及血管标志物白细胞分化抗原簇34(CD34)、血管内皮生长因子(VEGF)的表达。结果BONT/A促进SFI值恢复。髓鞘TB染色观测到BONT/A组在21d时坐骨神经恢复更显著。BONT/A能进一步增加p75NTR的表达,而对MPZ、BONT/A组在早期阶段其表达稍高,却在21d时显著降低MPZ的表达。BONT/A能随时间变化进一步促进血管标志物VEGF、CD34的表达。结论BONT/A对周围神经损伤后的神经再生有一定的促进作用,这种作用可能与增加损伤神经周围血管的生成,促进SCs的去分化和髓鞘再生有关。
Objective To explore the effects of botulinum toxin A(BONT/A)on sciatic nerve injury model,peripheral blood vessels,and schwann cells(SCs).Methods A total of 125 mice were randomly divided into a normal group(NC),control group(Control),and BONT/A group.The control group and BONT/A group(6U·kg^(-1))were divided into 7d group and 21d group,with 25 mice in each group.The sciatic nerve injury model was established in the control group and BONT/A group by the squeezing method,while the normal group was not treated.Observe the whole state of mice and evaluate their motor function with sciatic nerve function index(SFI).The regeneration and repair of the sciatic nerve were observed by toluidine blue(TB)staining.The expressions of p75 neurotrophin receptor(p75NTR)and myelin protein zero(MPZ)as well as vascular markers cluster of differentiation cluster 34(CD34)and vascular endothelial growth factor(VEGF)were detected by western blot.Results BONT/A promoted the recovery of SFI value.TB staining of the myelin sheath showed that the sciatic nerve recovery in the BONT/A group was more significant on the 21st day.BONT/A can further increase the expression of p75NTR,while the expression of MPZ and BONT/A group is slightly higher in the early stage,but it significantly reduces the expression of MPZ on the 21st day.BONT/A can further promote the expression of vascular markers VEGF and CD34 over time.Conclusions BONT/A can promote nerve regeneration after peripheral nerve injury,which may be related to increasing the formation of peripheral blood vessels and promoting the dedifferentiation and myelin regeneration of SCs.
