锡叶藤根总黄酮提取工艺优化及其抗炎活性分析

摘要 为了优化锡叶藤根中总黄酮的提取工艺,并对其抗炎活性展开探究,以总黄酮提取量作为衡量标准,运用单因素实验考察EtOH浓度、液料比、提取时间及温度对锡叶藤根总黄酮提取量的影响,基于此,运用正交实验对总黄酮提取条件予以优化;采用二甲苯致小鼠耳肿胀和角叉菜胶所致小鼠足肿胀建立急性炎症模型,以对锡叶藤根总黄酮的抗炎活性进行评价。结果表明锡叶藤根总黄酮最佳提取工艺为50%的EtOH,液料比为10∶1,在80℃的温度环境中,加热回流提取45 min,此条件下的总黄酮的得率最高值为233.5 mg/g。锡叶藤根总黄酮对小鼠耳肿胀有显著抗炎效果(P<0.01),能明显降低小鼠足肿胀程度(P<0.05)。该提取工艺简单又稳定,可被用于锡叶藤根总黄酮的提取,锡叶藤根总黄酮具有一定程度的抗炎活性。 In order to optimize the extraction process of total flavonoids from the Tetracera asiatica roots and explore their anti-inflammatory activity,a single-factor experiment was employed to investigate the effects of EtOH concentration,liquid-to-solid ratio,extraction time and temperature on total flavonoid yield from Tetracera asiatica roots.Based on this,orthogonal experiments were used to optimized the extraction conditions for total flavonoids.Acute inflammatory models were established in mice using xylene-induced ear swelling and carrageenan-induced paw swelling to evaluate the anti-inflammatory activity of total flavonoids from Tetracera asiatica roots.The results showed that the optimal extraction process of total flavonoids from Tetracera asiatica roots was as follows:50%ethanol,10∶1 liquid-to-solid ratio,and 80℃for 45 minutes of heating and refluxing,yielding the highest total flavonoid content of 233.5 mg/g.The total flavonoids extracts showed a remarkable anti-inflammatory effect on xylene-induced mouse ear swelling(P<0.01)and carrageenan-induced paw swelling(P<0.05).This extraction process of total flavonoids from Tetracera asiatica roots was simple,stable,and reliable,which possessed a certain degree of antiinflammatory activity.
