胶体金免疫层析法快速检测韭菜中肟菌酯残留

摘要 [目的]建立一种针对韭菜中肟菌酯残留的快速检测方法,并应用于实际样品检测,为农产品检测实验室提供快速筛查技术。[方法]优化前处理技术,将胶体金免疫层析技术应用于韭菜基质中肟菌酯的检测,验证方法的检出限、灵敏度、假阳性率、假阴性率和特异性,并通过液相色谱-串联质谱法确认方法的准确性。[结果]建立的胶体金免疫层析法对韭菜中肟菌酯的检出限为0.7 mg/kg,灵敏度100%,假阳性率3%,假阴性率0;与醚菌酯、吡唑醚菌酯、啶氧菌酯、嘧菌酯4种类似物无交叉反应;实际样本检测结果与液相色谱-串联质谱法结果符合性较好。[结论]本方法具有操作简便、检测速度快、准确性高等优点,适用于基层实验室的大批量样本检测及现场快速检测,对保障农产品质量安全具有现实意义。 [Aims]A rapid method for the determination of trifloxystrobin residues in leek was established and applied to the detection of actual samples to provide a rapid screening technique for testing laboratory of agricultural products.[Methods]By optimizing the pretreatment technique,colloidal gold immunochromatographic assay(GICA)was applied to the detection of trifloxystrobin in leek matrix.The detection limit,sensitivity,false positive rate,false negative rate and specificity of the method was verified,and the accuracy was confirmed by liquid chromatography-tandem mass spectrometry(LC-MS/MS).[Results]The detection limit of trifloxystrobin in leek by GICA was 0.7 mg/kg,with the sensitivity of 100%,the false positive rate of 3%,and the false negative rate of 0.There was no cross-reaction with its four analogues kresoxim-methyl,pyraclostrobin,picoxystrobin and azoxystrobin.The results of actual samples were in good agreement with those of LC-MS/MS.[Conclusions]The method has the advantages of simple operation,fast detection speed and high accuracy,which is suitable for the mass sample detection in the basic laboratory and field rapid detection,and has practical significance for ensuring the quality and safety of agricultural products.
