摘要
目的:研究苍术标准汤剂中苍术素和绿原酸的量值传递规律,为苍术配方颗粒及其复方制剂质量标准制定提供参考依据。方法:以苍术素、绿原酸为指标成分,结合出膏率、指纹图谱相似度、成分转移率、指标成分含量,采用HPLC法建立饮片与标准汤剂指纹图谱,对苍术饮片向标准汤剂量值传递规律进行含量测定。苍术标准汤剂指纹图谱标定8个共有峰,其中5个共有峰传递到标准汤剂上,并指认共有成分苍术素、绿原酸2个;以峰面积聚类分析标准汤剂,结果与指纹图谱相似度呈一致规律。含量测定结果显示,标准汤剂出膏率为21.26%~29.97%;苍术素含量为0.570%~1.283%,转移率为0.04%~0.25%;绿原酸含量为0.033%~0.223%,转移率为31.09%~67.41%。结论:结果显示,苍术标准汤剂中绿原酸的转移率较高,而药典指标成分苍术素的转移率极低,表明绿原酸可作为苍术标准汤剂质量控制成分之一,而苍术素作为标准汤剂质量控制的指标成分有待进一步研究商榷,但该HPLC法稳定可靠,可为苍术标准汤剂的质量控制提供参考。
Objective:To study the quantitative transfer law of atractylodin and chlo-rogenic acid in Atractylodis rhizoma standard decoction,and to provide reference for establishing the quality standard of Atractylodis Rhizoma formula granules and its compound preparations.Methods:Taking atractylodin and chlo-rogenic acid as index components,combined with the extraction rate,fingerprint similarity,component transfer rate and index component content,the fingerprint of Atractylodis rhizoma pieces and standard decoction was established by HPLC,and the transfer law of value from Atractylodis rhizoma pieces to standard decoction was determined.The fingerprint of Atractylodis rhizoma standard decoction demarcates 8 common peaks,among which 5 common peaks were transmitted to the standard decoction,and 2 common components,namely atractylodin and chlo-rogenic acid,were identified.According to the peak area cluster analysis of standard decoction,which was consistent with the similarity of fingerprint.The results of content determination showed that the extraction rate of standard decoction ranged from 21.26%to 29.97%.The content of atractylodin ranged from 0.570%to 1.283%,and the transfer rate was 0.04%to 0.25%.The content of chlo-rogenic acid ranged from 0.033%to 0.223%,and the transfer rate was 31.09%to 67.41%.Conclusion:The results show that the transfer rate of chlo-rogenic acid in the standard decoction of Atractylodis rhizoma is high,but the transfer rate of atractylodin in Pharmacopoeia is extremely low,which indicates that chlo-rogenic acid can be used as one of the quality control components of the standard decoction of Atractylodis rhizoma,and atractyl as the index component of the quality control of the standard decoction needs further study and discussion.However,the HPLC method is stable and reliable,which can provide reference for the quality control of Atractylodis rhizoma standard decoction.
