基于AGE-RAGE/p38MAPK信号通路探讨温阳复元方对脑缺血再灌注损伤大鼠脑损伤的保护作用

Protective effects of Wenyang Fuyuan Recipe on cerebral ischemia-reperfusion injury in rats via AGE-RAGE/p38MAPK signaling pathway

摘要 目的探究温阳复元方对脑缺血再灌注损伤(CIRI)大鼠脑损伤的保护作用。方法大鼠随机分为假手术组、模型组、FPS-ZM1组、温阳复元方组,每组6只。除假手术组外,其余各组大鼠建立CIRI模型,采用TTC染色和改良神经病学严重程度评分(mNSS)评判模型成功与否,TUNEL染色检测神经细胞凋亡情况,免疫荧光染色检测脑组织RAGE、S100β蛋白表达,RT-qPCR法检测脑组织RAGE、p38MAPK mRNA表达,Western blot法检测脑组织RAGE、AGE、HMGB1、p38、Bcl-2蛋白表达。结果与假手术组比较,模型组大鼠脑组织出现明显白色梗死灶,梗死面积增加(P<0.05),神经功能评分升高(P<0.05),神经元凋亡细胞数增多(P<0.05),脑组织RAGE、S100β蛋白表达升高(P<0.05),脑组织AGE、RAGE、HMGB1、p38MAPK蛋白与mRNA表达升高(P<0.05);与模型组比较,FPS-ZM1组和温阳复元方组上述指标均有改善(P<0.05),其中温阳复元方组作用更佳。结论温阳复元方可能通过抑制AGE-RAGE及其下游p38MAPK信号通路的活化,从而减轻神经元凋亡,发挥对大鼠的脑保护作用。 AIM To explore the protective effect of Wenyang Fuyuan Recipe on cerebral ischemia-reperfusion injury(CIRI)in rats.METHODS Among the rats randomly divided into the sham operation group,the model group,the FPS-ZM1 group and Wenyang Fuyuan Recipe group,with 6 rats in each group,all were induced into the rat CIRI models except those of the sham operation group.The rats had the model success verified by TTC staining and modified neurological severity score(mNSS);their neuronal apoptosis detected by TUNEL staining;their cerebral protein expressions of RAGE and S100βdetected by immunofluorescence staining;their cerebral mRNA expressions of RAGE and p38MAPK detected by RT-qPCR;and their protein expressions of RAGE,AGE,HMGB1,p38 and Bcl-2 in brain tissue detected by Western blot.RESULTS Compared with the sham operation group,the model group showed marked white matter with an enlarged size in the brain tissue(P<0.05),increased neurological function score(P<0.05),increased number of apoptotic cells in neurons(P<0.05),increased cerebral protein expressions of RAGE and S100β(P<0.05),and increased protein and mRNA expressions of cerebral AGE,RAGE,HMGB1 and p38MAPK(P<0.05).Compared with the model group,the group intervened with either FPS-ZM1 or Wenyang Fuyuan Recipe shared improved conditions in terms of the aforementioned indicators levels(P<0.05),and Wenyang Fuyuan Recipe group showed its particular priority in the efficacy.CONCLUSION Wenyang Fuyuan Recipe may protect the rat brain via AGE-RAGE and its downstream p38MAPK signaling pathway mediated neuronal apoptosis attenuation.
